小儿咳喘灵颗粒HPLC指纹图谱的建立及化学模式识别研究

摘要 目的建立小儿咳喘灵颗粒的化学成分指纹图谱,并应用化学模式识别进行分析。方法采用高效液相色谱法(HPLC)分离小儿咳喘灵颗粒的主要组分,利用药典委员会的指纹图谱相似度评价系统(2012版),建立小儿咳喘灵颗粒的指纹图谱并进行相似度评价,通过与对照品比对进行色谱峰的指认和归属分析,分别采用SPSS 25.0、SIMCA 14.1软件进行聚类分析、主成分分析。结果10批小儿咳喘灵颗粒HPLC指纹图谱中有16个共有峰,与对照图谱的相似度在0.901~0.962之间,通过聚类分析将10批样品分为两类,主成分分析与聚类分析结果一致。通过与对照品图谱比对,确定了9个已知峰,分别为苦杏仁苷、4,5-O-二咖啡酰奎宁酸、木犀草苷、盐酸麻黄碱、R,S-告依春、绿原酸、甘草苷、3,5-O-二咖啡酰奎宁酸和甘草酸铵。结论建立的指纹图谱方法结合化学模式识别分析可较好地评价小儿咳喘灵颗粒质量。 Objective To establish the HPLC fingerprints of xiaoer kechuanling granules and evaluate its quality with chemical pattern recognition method.Methods Separation of main components of xiaoer kechuanling granules by HPLC.Using the similarity evaluation system of chromatographic fingerprint of traditional Chinese medicine(2012 version),establish the fingerprint of xiaoer kechuanling granules.Identification and attribution analysis of chromatographic peaks were carried out by comparison with the chromatograms of the reference substance.SPSS 25.0 and SIMCA 14.1 software were used for cluster analysis and principal component analysis.Results There were 16 common peaks in high performance liquid chromatography fingerprint of 10 batches of xiaoer kechuanling granules.The similarity with the control atlas was between 0.901 and 0.962.The 10 batches of samples were divided into two categories by cluster analysis,and the results of principal component analysis were consistent with those of cluster analysis.9 common peaks were determined by comparison with the control atlas.They were amygdalin,4,5-O-dicaffeoyl quinic acid,luteolin,ephedrine hydrochloride,R,S-hydantoin,chlorogenic acid,glycyrrhizin,3,5-O-dicaffeoyl quinic acid and ammonium glycyrrhizinate.Conclusion The established fingerprint method with chemical pattern recognition analysis can be used for the quality evaluation of xiaoer kechuanling granules.
