摘要
目的 探究山姜素对大鼠妊娠高血压的作用及可能机制。方法 雌雄大鼠以2∶1合笼饲养,妊娠后采用随机数字表法将孕鼠分为对照组(Control组)、妊娠高血压组(PIH组)和山姜素组(Alpinetin组),每组10只。测量大鼠血压和24 h尿蛋白量;HE染色观察胎盘组织形态学变化;免疫荧光检测胎盘组织ET-1蛋白表达;qRT-PCR检测胎盘组织PPAR-γ和ETAR mRNA表达;Western blot检测胎盘组织PPAR-γ和ETAR蛋白表达。结果 山姜素降低妊娠高血压大鼠血压和24 h尿蛋白量,改善胎盘组织形态,降低胎盘组织ET-1和ETAR表达,增加胎盘组织PPAR-γ表达。结论 山姜素改善大鼠妊娠高血压,其作用机制可能与激活PPAR-γ,抑制ET-1/ETAR信号通路有关。
Objective To explore the therapeutic effect and possible mechanism of alpinetin on pregnancy hypertension in rats.Methods Female and male rats were fed in cages 2:1.After pregnancy,the pregnant rats were divided into Control group,PIH group and Alpinetin group by random number table method,with 10 rats in each group.Blood pressure and 24 h urinary protein were measured.The morphological changes of placenta were observed by HE staining.The expressions of ET-1 protein in placenta were detected by immunohistochemical staining.The levels of PPAR-mRNA and ETAR mRNA in placenta were detected by qRT-PCR.The expressions of PPAR-and ETAR in placenta were detected by Western blot.Results Alpinetin decreased blood pressure and 24 h urinary protein content,improved the morphology of placental tissue,decreased the expressions of ET-1 and ETAR in placental tissue,and increased the expression of PPAR-in placental tissue in pregnant hypertensive rats.Conclusion The effect of alpinetin on pregnancy hypertension in rats may be related to the activation of PPAR-and the inhibition of ET-1/ETAR signaling pathway.
