GPC3通过PI3K/AKT通路参与肺鳞状细胞癌的发生发展 被引量:1

摘要 目的 通过生物信息学分析GPC3在肺鳞状细胞癌(LUSC)中的差异表达情况并探讨GPC3对LUSC细胞增殖的影响。方法 从GEO数据库下载LUSC的表达谱数据,采用R-studio分析差异表达基因,通过免疫组化方法检测GPC3在LUSC组织和癌旁正常组织的表达情况。构建稳定GPC3高表达及沉默的NCI-H226细胞系,CCK-8法和克隆形成实验检测GPC3对LUSC细胞增殖的影响;Western blot法检测GPC3变化对PI3K/AKT信号通路关键蛋白的影响。结果 GPC3在LUSC中存在差异表达(P<0.01);免疫组化结果发现GPC3在正常组织中不表达,在LUSC组织中表达率为70%。GPC3高表达可促进LUSC细胞的增殖,上调PI3K及pAkt表达。结论 GPC3可能通过调节PI3K/Akt信号通路促进LUSC的细胞增殖。 Objective The differential expression of GPC3 in lung squamous cell carcinoma(LUSC)was analyzed by bioinformatics and the effect of GPC3 on the proliferation of LUSC cells was investigated.Methods The expression profile data of LUSC were downloaded from GEO database,the differentially expressed genes were analyzed by R-studio,and the expression of GPC3 in LUSC tissues and adjacent normal tissues was detected by immunohistochemical method.A stable NCI-H226 cell line with high expression and silence of GPC3 was constructed,and the effect of GPC3 on the proliferation of LUSC cells was detected by CCK-8 assay and clonal formation assay.The effects of CPC3 changes on key proteins of PI3K/AKT signaling pathway were detected by Western blot.Results GPC3 was differentially expressed in LUSC(P<0.01).The immunohistochemical results showed that GPC3 was not expressed in normal tissues,and the rate of positive expression in LUSC tissues was 70%.The high expression of GPC3 can promote the proliferation of LUSC cells and up-regulate the expressions of PI3K and pAkt.Conclusion GPC3 may promote LUSC cell proliferation by regulating the PI3K/Akt signaling pathway.
