摘要
目的:筛选不同产地金银花药材特征性组分,为其产地鉴别及质量评价提供科学依据。方法:采集3个不同产区(山东省、河南省、河北省)55批金银花药材的超高效液相色谱-三重四极杆串联质谱(UPLC-QQQMS/MS),对绿原酸、木犀草苷、咖啡酸等14个指标性成分进行定量检测,结合聚类分析、偏最小二乘判别分析等模式识别方法进行质量评价。结果:不同产地金银花样品共标定14个共有峰,其中河南省新乡产区药材中獐芽菜苦苷含量最高,河北省金银花中木犀草素成分含量最高。各批样品聚为3类,木犀草苷、金丝桃苷、咖啡酸、木犀草素、异绿原酸B、断氧化马钱子苷、1,5-二咖啡酰奎宁酸为质量差异标志物。结论:UPLC-QQQ-MS/MS多成分定量结合模式识别的方法能科学评价不同产地金银花药材的质量。
Objective:The characteristic components of Lonicerae Japonicae Flos were screened to provide a scientific basis for the identification and quality evaluation of Lonicerae Japonicae Flos from different regions.Methods:Ultra-performance liquid chromatography coupled with a triple quadrupole mass spectrometry(UPLC-QQQ-MS/MS)of 55 batches of Lonicerae Japonicae Flos samples from three different producing areas(Shandong province,Henan province and Hebei province)were collected.At the same time,a total of fourteen components including chlorogenic acid,luteolin-7-O-glucoside and caffeic acid were quantitatively analyzed.The pattern recognition methods such as cluster analysis and partial least squares discriminant analysis were used to evaluate quality.Results:There were fourteen common peaks in the fingerprints of 55 batches of Lonicerae Japonicae Flos samples.The content of swertimarin in Xinxiang,Henan province was the highest,and the content of luteolin from Hebei province was the highest.Various batches of samples were clustered into three categories.Seven components,including luteoloside,hyperoside,caffeic acid,luteolin,isochlorogenic acid B,secoxyloganin,1,5-Dicaffeoylquinic acid,were screened by partial least squares discriminant analysis as Q-markers of Lonicerae Japonicae Flos.Conclusion:UPLC-QQQ-MS/MS multi-component determination combined with pattern recognition methods can evaluate the quality of Lonicerae Japonicae Flos from different producing areas scientifically.
