摘要
目的通过弗氏完全佐剂(complete Freund’s adjuvant,CFA)建立自身免疫性慢性非细菌性前列腺炎(chronic nonbacterial prostatitis,CNP)大鼠模型,观察其对前列腺病理组织学及相关促炎细胞因子表达水平的影响,为CNP的研究提供参考。方法48只Wistar大鼠随机分为空白组、假手术组、CFA原位注射组、CFA与大鼠前列腺蛋白混合物原位注射组,造模后28 d处死,取大鼠前列腺组织。称重并计算各组大鼠前列腺指数,HE染色观察各组大鼠前列腺组织病理损伤情况,ELISA检测大鼠前列腺组织促炎细胞因子IL-1β、IL-6、COX-2水平。结果假手术组与空白组相比,大鼠前列腺指数无显著差异,前列腺组织无明显病理改变,前列腺组织IL-1β、IL-6、COX-2无显著变化。与假手术组相比,CFA原位注射组与混合物原位注射组均出现前列腺指数显著增高,前列腺组织出现明显病理改变与炎性细胞浸润,前列腺组织IL-1β、IL-6、COX-2表达显著增加。混合物原位注射组与CFA原位注射组相比,出现更显著的前列腺组织炎性细胞浸润,且前列腺组织IL-1β、IL-6表达水平显著增高。结论CFA原位注射及CFA与大鼠前列腺蛋白混合物原位注射方法均可成功制备CNP大鼠模型,可为CNP的进一步研究提供模型基础。
Objective To establish the chronic nonbacterial prostatitis(CNP)autoimmune rat models by complete Freund's adjuvant(CFA),and observe its effect on prostate pathological tissue and related proinflammatory cytokines,to provide reference for CNP research.Methods sForty-eight Wistar rats were randomly divided into four groups such as the blank group,the sham group,CFA in situ injection group,and CFA and rat prostate protein mixture in situ injection group.After 28 days of modeling,the rats were euthanized and their prostate tissues were collected.The prostate index of each group was calculated.HE staining was used to observe the damage of prostate tissue in each group.ELISA was used to detect the levels of proinflammatory cytokines IL-1β,IL-6 and COX-2 in prostate tissue.Results There were no significant differences in prostate index,prostate tissue damage,and the level IL-1β,IL-6 and COX-2 in prostate tissue between the blank group and the sham group.Compared with the rats in the sham group,the rats in the CFA groups showed the higher prostate index,more obvious prostate tissue damage and inflammatory cell infiltration,the higher IL-1β,IL-6 and COX-2 expression in the prostate tissue.Compared with rats in CFA in situ injection group,the rats in the mixture in situ injection group showed more obvious inflammatory cell infiltration and significantly increased IL-1βand IL-6 levels in prostate tissue.Conclusion Both CFA in situ injection and CFA and rat prostate protein mixture in situ injection could successfully form CNP rat models,which can provide the model basis for CNP study.
