多指标综合评分法优化养阴生肌肠溶胶囊水提醇沉工艺

Optimization of water extraction and alcohol precipitation process of Yangyin Shengji enteric-coated capsule by multi-index comprehensive scoring method

优化养阴生肌肠溶胶囊水提醇沉工艺。采用星点设计-效应面法,以浸泡时间、提取时间、加水量、提取次数为考察因素,以龙胆苦苷、芍药苷、甘草苷、盐酸小檗碱、6-姜酚、吴茱萸碱、吴茱萸次碱的含量及干膏得率为评价指标,利用熵权法进行多指标综合评分,优化水提工艺;采用L9(34)正交试验法,以浓缩比、醇沉浓度、静置时间为考察因素,以龙胆苦苷、芍药苷、甘草苷、盐酸小檗碱、6-姜酚、吴茱萸碱、吴茱萸次碱的含量及干膏得率为评价指标,利用熵权法进行多指标综合评分,优化醇沉工艺。结果表明,优化的水提工艺为加12倍量水,浸泡45min,提取2次,每次90min;优化的醇沉工艺为浓缩比1∶2,醇沉浓度50%,静置30h。本研究优选的水提醇沉工艺稳定性好、重复性高,可为养阴生肌肠溶胶囊后续的成型工艺研究提供参考。

To optimize the water extraction and alcohol precipitation process of Yangyin Shengji enteric-coated capsules.The central composite design-response surface method was used to optimize the water extraction process.The soaking time,extraction time,water addition and extraction times were used as the investigation factors,and the contents of gentiopicroside,paeoniflorin,liquiritin,berberine hydrochloride,6-gingerol,evodiamine,rutaecarpine and dry extract yield were used as the investigation indexes.The entropy method was used for multi-index comprehensive scoring to optimize the water extraction process.L9(34)orthogonal test method was used to investigate the concentration ratio,the concentration of alcohol precipitation and the standing time.The contents of gentiopicroside,paeoniflorin,liquiritin,berberine hydrochloride,6-gingerol,evodiamine,rutaecarpine and the yield of dry extract were used as the indexes.The entropy weight method was used for multi-index comprehensive scoring to optimize the alcohol precipitation process.The results showed that the optimized water extraction process was adding 12 times the amount of water,soaking for 45 min,extracting for 2 times,90 min each time;the optimized alcohol precipitation process was a concentration ratio of 1∶2,the alcohol precipitation concentration was 50%,and the standing time was 30 h.The optimized water extraction and alcohol precipitation process in this study has good stability and high repeatability,which can provide reference for the subsequent molding process research of Yangyin Shengji enteric-coated capsules.
