液体培养甘薯组培苗对培养基分装量和接种茎段的优化 被引量:1

Optimization of culture-medium portion and inoculated stem segment for tissue culture of liquid sweet potato seedlings

摘要 为降低甘薯组培苗生产成本,提高快繁效率,以甘薯脱毒组培苗茎段为外植体,以MS+IBA(0.1 mg/L)+蔗糖(30 g/L)为扩繁培养基,文章研究了培养方式、培养基分装量及接种茎段长度与形态对甘薯组培苗生长的影响.结果表明:液体培养优于同等培养条件下的固体培养和玻璃棉支撑培养.采用液体培养快繁甘薯组培苗,以30 mL/瓶的扩繁培养基分装量最有利于组培苗的生长与增殖,保留半截叶柄且长度为1.5~2.0 cm的茎段是最适宜的外植体,不仅便于切割与接种,且成苗率高,组培苗生长茁壮、迅速.因此,甘薯脱毒苗最佳的离体快繁方法是截取1.5~2.0 cm长并保留半截叶柄的茎段,接种于30 mL/瓶液体培养基,经过一个培养周期的离体培养,不仅茎段腋芽萌发不受影响,成苗率与成苗数高,且组培苗生长迅速,增殖系数高. In order to reduce the production cost and improve the rapid propagation efficiency of sweet potato tissue culture seedlings,the effects of culture methods,media content,length and shape of inoculated stem segments on the growth of sweet potato tissue culture seedlings were studied by using different stem segments of virus-free tissue culture plantlet of sweet potato as explants and cultured on propagation medium supplemented with 0.1 mg/L IBA(3-Indolebutyric acid)+30 g/L sucrose.The results showed that liquid culture was superior to solid culture and liquid culture using aluminum silicate wool as supports under the same culture conditions.When using liquid culture for rapid propagation of sweet potato tissue culture seedlings,the content volume of 30 mL media per culture flask was most conducive to the growth and proliferation of tissue culture seedlings.The stem segment with half petiole and length of 1.5~2.0 cm were the most appropriate explants for rapid propagation of sweet potato seedlings,which were not only convenient for cutting and inoculation of explants,but also had a high seedling rate and rapid growth of tissue culture seedlings.Therefore,the best protocol for rapid propagation of virus-free sweet potato seedlings in vitro was to inoculate stem segments with half petiole retained and length of 1.5~2.0 cm in 30mLl liquid media for one growth period of culture,and,as a result,not only the axillary bud germination of stem segments was not affected,but also the rate and number of seedlings were high,and the tissue culture seedlings grew rapidly with high proliferation coefficient.
