摘要
为获得具有高抗氧化活性的黄芩提取物,本试验对比水提和醇提两种提取方式,并将提取物冻干后复溶成水提水溶、醇提醇溶、醇提水溶三种状态,以胃肠模拟消化后产物的DPPH自由基清除率、总抗氧化能力、·OH自由基清除率、总还原能力和超氧阴离子清除率五种抗氧化活性指标的综合秩和比(RSR值)为判断标准,确定出最佳的提取溶剂。并在此基础上采用单因素试验初步确定其提取工艺的主要影响因素及水平,再结合析因设计试验分析提取工艺各因素之间的交互作用,最终获得最佳的提取工艺参数。通过RSR值排序的结果表明:两种冻干粉在复溶后的抗氧化活性从大到小的顺序为:水提水溶、醇提水溶、醇提醇溶。单因素试验最佳提取工艺为:温度保持微沸,提取时间120 min,液料比50:1(mL/g)。析因设计试验最佳提取工艺为:温度保持微沸,提取时间120 min,液料比60:1(mL/g)。优选的工艺提取液其经过消化后DPPH自由基清除率为39.48%、ABTS+自由基清除率为58.85%、·OH自由基清除率为48.63%、总还原能力为0.49、超氧阴离子清除率为28.87%,可为黄芩抗氧化活性物质的提取提供重要参考依据。
In order to obtain Scutellaria baicalensis extract with high antioxidant activity,the comparison of two methods(water extract and alcohol extract)on Scutellaria baicalensis was conducted,and the freeze-dried powder of Scutellaria baicalensis extract was redissolved into three states:water dissolved water extract,alcohol dissolved alcohol extract and water dissolved alcohol extract.The comprehensive rank-sum ratio(RSR)of the five indexes which were DPPH free radical clearance rate,total antioxidant capacity,·OH free radical clearance rate and total reduction ability and superoxide anion scavenging rate of the Scutellaria baicalensis extract after gastrointestinal simulated digestion was set as judgment standard to select optimized extract solvent.The single factor experiment was used to preliminarily determine the main influencing factors and level of the extraction process under optimized extract solvent.Then factorial design experiment was used to analyze the interaction between the factors of the extraction process,so as to obtain the best extraction process parameters.The results of RSR value sequencing showed that the antioxidant activity of the two lyophilized powders after redissolution was in the order of water extraction and water dissolution,alcohol extraction and water dissolution and alcohol extraction and alcohol dissolution.The results of single factor experiment showed that the extraction temperature was boiling water,the extraction time was 120 min,and the liquid-material ratio was 50:1(mL/g).In factorial design the experimental results were as follows:the extraction temperature was boiling water,the extraction time was 120 min,and the liquid-material ratio was 60:1(mL/g).After digestion,the DPPH free radical scavenging rate,ABTS+free radical scavenging rate,·OH free radical scavenging rate,total reducing capacity and superoxide anion scavenging rate of the optimized process extract were 39.48%,58.85%,48.63%,0.49 and 28.87%respectively,It could provide important reference basis for the extraction of antioxidant active substances from Scutellaria baicalensis.
