摘要
目的大脑皮层厚度是重性抑郁障碍(MDD)患者大脑结构性改变的重要方面,并可能反映疾病背后的病理生理机制。该研究旨在采用基于微分配准的皮质厚度算法探究初诊MDD患者大脑皮层厚度变化。方法该研究招募了17例初诊未用药的MDD患者和17名健康志愿者,并收集了所有受试者的人口统计学资料,并在确定入组后的2周之内完成全脑T1结构像MRI扫描,经分割、配准和标准化后获得全脑皮层厚度。皮层厚度的组间比较采用置换检验和FDR多重比较校正。结果MDD组和对照组的皮层厚度经多重比较校正后P值均大于0.05,差异无统计学意义。基于微分配准的皮质厚度计算方法不能区分MDD与对照组。结论相较于既往研究报告的MDD与皮质厚度变化之间的关联,该研究中发现这种关联是相对较弱的,未来需要在更大的样本中验证皮层厚度变化的稳定性。
Objective Cortical thickness is a crucial component of structural brain changes in major depressive disorder(MDD)patients and might be a reflection of the pathophysiological processes that underlie the disease.This study used a diffeomorphic registration-based cortical thickness measurement to examine cortical thickness changes in patients with drug-naive MDD.Methods 17 patients with drug-naive MDD and 17 healthy volunteers were recruited,and demographic data were gathered on each participant.Within two weeks after the enrollment decision,the whole-brain T1 structural image MRI scans were also completed,and after segmentation,alignment,and standardization,the whole-brain cortical thickness had been estimated.Intergroup comparisons of cortical thickness were made using a permutation test and FDR multiple comparison correction.Results The cortical thicknesses of the MDD group and control group were corrected for multiple comparisons with P-values greater than 0.05,and the differences were not statistically significant.The method of diffeomorphic registration based cortical thickness could not differentiate MDD with healthy controls.Conclusion This association was found to be relatively weak in the present study compared to the association between MDD and cortical thickness change reported in earlier studies,and the stability of cortical thickness change needs to be confirmed in a larger sample in the future.
