摘要
目的探讨血流限制训练联合富血小板血浆(PRP)对兔骨关节炎(OA)模型软骨修复的作用。方法50只3月龄雄性新西兰纯种兔,按数字表法分为5组,正常对照组、OA模型组、PRP治疗组、血流限制组、PRP+血流限制组,每组10只,均干预6周后,采用HE染色观察关节软骨组织病理变化,RT-PCR技术分析Ⅱ型胶原(typeⅡcollagen,ColⅡ)、蛋白多糖(proteoglycan,PG)及基质金属蛋白酶13(matrix metalloproteinase 13,MMP13)mRNA的表达。结果通过HE染色直接观察膝关节软骨细胞数量及排列,发现OA模型组关节软骨表面出现不同程度损伤;与OA模型组的改良Minkin病理评分比较,PRP治疗组、血流限制组、PRP+血流限制组关节软骨表面损伤均减轻(P<0.05);PRP+血流限制组的形态改变又明显好于PRP治疗组和血流限制组(P<0.05)。OA模型组ColⅡ、PG的mRNA相对表达量明显小于正常对照组(P<0.05),MMP13表达量高于正常对照组(P<0.05);PRP治疗组、血流限制组、PRP+血流限制组ColⅡ、PG表达均高于OA模型组(P<0.05),同时MMP13表达减少(P<0.05)。结论血流限制训练联合PRP可以促进兔骨关节炎模型软骨修复。
Objective To investigate the effect of blood flow restriction training combined with platelet�rich plasma(PRP)on cartilage repair in a rabbit osteoarthritis(OA)model.Methods Fifty 3-month-old male New Zealand purebred rabbits were divided into five groups according to the numerical table method:the control group,the OA model group,the PRP treatment group,the blood flow restriction group,and PRP+blood flow restriction group,with 10 rabbits in each group.Rabbits in all groups were intervened for six weeks,and then the histopathological changes of articular cartilage were observed by HE staining,and the mRNA expression of typeⅡcollagen(ColⅡ),proteoglycan(PG)and matrix metalloproteinase 13(MMP13)were analyzed by RT-PCR.Results The number and arrangement of cells in the knee cartilage were directly observed by HE staining,and the surface of articular cartilage in the OA model group showed different degrees of damage;compared with the modified Minkin pathology score in the OA model group,the damage to the articular cartilage surface was reduced in the PRP-treated group,the blood flow-restricted group,and the PRP+blood flow-restricted group(P<0.05);and the morphology of the PRP+blood flow-restricted group was significantly better than that of the PRP-treated group and the blood flow-restricted group(P<0.05).The relative mRNA expression of ColⅡand PG in the OA model group was significantly lower than that in the normal control group(P<0.05),and the expression of MMP13 was higher than that in the normal control group(P<0.05),and the expression of ColⅡand PG was higher in the PRP treatment group,the blood flow restriction group,and the PRP+blood flow restriction group than that in the OA model group(P<0.05).Meanwhile,the MMP13 expression was reduced(P<0.05).Conclusion Blood flow restriction training combined with PRP can promote cartilage repair in rabbit osteoarthritis model.
