Optimization of Colloid Mill Inclusion Process of Lianqiao(Forsythia Suspensa)Volatile Oil Based on Box-Behnken Response Surface Methodology

摘要 目的:优化连翘挥发油胶体磨包合工艺。方法:以连翘挥发油的包合率和包合物收率及综合评分为指标比较饱和水溶液法、超声法、胶体磨研磨法对连翘挥发油的包合效果。在单因素试验的基础上,采用Box-Behnken响应面法进行优化,以挥发油(mL)与β-环糊精(g)的投料比、β-环糊精(g)与水的投料比和研磨时间为考察因素,确定连翘挥发油胶体磨包合的最佳工艺条件。结果:胶体磨研磨法的包合率、包合物得率及综合评分均高于饱和水溶液法和超声法。最佳的包合工艺为挥发油与β-环糊精的投料比为1∶10,β-环糊精与水的投料比为1∶6,研磨时间为20 min。3次试验结果显示,包合率平均值为89.52%,包含物得率平均值为93.98%,综合得分为90.41%,RSD为1.21%。结论:优选的连翘挥发油胶体磨包合工艺操作简便,包合效果好,适合大规模工业化生产。 Objective:To optimize the inclusion process of volatile oil from Lianqiao(Forsythia Suspensa)by colloid mill.Methods:The inclusion rate,inclusion compound yield and comprehensive score of Lianqiao(Forsythia Suspensa)volatile oil were used as indexes to compare the inclusion effect of saturated aqueous solution method,ultrasonic method and colloid mill grinding method on Lianqiao(Forsythia Suspensa)volatile oil.On the basis of single factor experiment,the Box-Behnken response surface method was used to optimize the process.The ratio of volatile oil(mL)toβ-cyclodextrin(g),the ratio ofβ-cyclodextrin(g)to water and the grinding time were used as the investigation factors to determine the optimum conditions for the inclusion of Lianqiao(Forsythia Suspensa)volatile oil by colloid mill.Results:The inclusion rate,inclusion compound yield and comprehensive score of colloid mill grinding method were higher than those of saturated aqueous solution method and ultrasonic method.The optimum inclusion process was as follows:the ratio of volatile oil toβ-cyclodextrin was 1∶10,the ratio ofβ-cyclodextrin to water was 1∶6,and the grinding time was 20 min.The results of three experiments showed that the average inclusion rate was 89.52%,the average inclusion rate was 93.98%,the comprehensive score was 90.41%,and the RSD was 1.21%.Conclusion:The optimized colloid mill inclusion process of volatile oil from Lianqiao(Forsythia Suspensa)is simple and easy to operate,and the inclusion effect is good,which is suitable for large-scale industrial production.
