摘要
目的:探讨针刀联合葛根素膝关节腔内注射对兔膝骨关节炎模型的影响。方法:选择新西兰白兔30只,随机分为空白对照组、模型组、葛根素组、针刀联合葛根素组和针刀联合透明质酸钠组,每组6只。除空白对照组外,其余4组均制备兔膝骨关节炎动物模型。造模成功后,葛根素组关节腔内注入葛根素注射液,针刀联合葛根素组针刀松解术的同时注入葛根素注射液,针刀联合透明质酸钠组予以针刀松解术的同时注入透明质酸钠,空白对照组和模型组注射等体积的生理盐水,各组均连续治疗5周。RT-PCR检测膝兔软骨检测基质金属酶蛋白1(matrix metalloproteinases-1,MMP-1)和基质金属酶蛋白13(matrix metalloproteinases-1,MMP-13)的mRNA含量,ELISA法检测兔关节腔内细胞因子肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)和白细胞介素1(interleukin-1,IL-1)、白细胞介素6(Interleukin-1,IL-6)的含量。结果:针刀联合葛根素组中IL-1、IL-6和TNF-α水平及MMP-1和MMP-13的mRNA表达较其他组明显减少(P<0.05)。结论:针刀联合葛根素可以明显减少软骨关节肿MMP-1和MMP-13的mRNA含量,降低IL-1、IL-6和TNF-α细胞因子表达。
Objective:To explore the influence of needle-knife and intra-articular injection of puerarin injection on rabbit models suffering knee osteoarthritis(KOA).Methods:All 30 New Zealand rabbits were randomized into blank control group,the model group,puerarin group,needle-knife combined with puerarin group,needle-knife and sodium hyaluronate group,with six ones in each group.Except for the blank control group,the remaining four groups were prepared into KOA rabbit models.After successful modeling,puerarin group accepted intra-articular injection of puerarin injection,needle-knife combined with puerarin group needleknife relaxation and puerarin injection,blank control group and the model group were injected with equal volume of physiological saline,consecutively treated for five weeks.RT-PCR was used to detect the contents of MMP-1 and MMP-13 mRNA in rabbit knee cartilage,ELISA method to detect the expressions of TNF-α,IL-1 and IL-6 in knee articular cavity.Results:The expressions of IL-1,IL-6 and TNF-α,MMP-1 and MMP-13 mRNA in needleknife combined with puerarin group reduced remarkably than the other groups(P<0.05).Conclusion:Needle-knife and intra-articular injection of puerarin injection could apparently lower the contents of MMP-1 and MMP-13 mRNA in knee cartilage and the expressions of IL-1,IL-6 and TNF-α.
