胡桃醌对肺癌NCL-H460细胞凋亡机制的影响

The effect of juglone on apoptosis mechanism of lung cancer NCL-H460 cells

摘要 目的观察胡桃醌对肺癌NCL-H460细胞凋亡机制的影响。方法应用不同浓度胡桃醌作用于肺癌NCL-H460细胞,用流式细胞术检测细胞凋亡情况,用Western Blot法检测Bcl-2、Bax、p-p53蛋白表达水平。结果胡桃醌促进肺癌NCL-H460细胞凋亡,并存在剂量效应;与对照组相比,实验组Bcl-2表达水平降低,Bax、p-p53表达水平显著升高(P<0.05)。结论胡桃醌可通过P53途径诱导肺癌NCL-H460细胞凋亡。 Objective To observe the effect of Juglone on the apoptosis mechanism of lung cancer NCL-H460 cells.Methods Different concentrations of Juglone were applied to treat lung cancer NCL-H460 cells.The apoptosis was detected by flow cytometry,and the expression levels of Bcl-2,Bax,and p-p53 were detected by Western Blot.Results Juglone promotes apoptosis of lung cancer NCL-H460 cells and has a dose effect;Compared with the control group,the expression level of Bcl-2 in the experimental group was decreased,and the expression level of Bax,p-p53 was significantly increased(P<0.05).Conclusion Juglone can induce apoptosis of lung cancer NCL-H460 cells through P53 pathway.
