宫颈癌患者miR-34a、miR-145、miR-21和Ki-67的表达及其与HPV感染的关系 被引量:1

Expressions of miR-34a,miR-145,miR-21 and Ki-67 in cervical cancer patients and their association with HPV infection

摘要 目的分析宫颈癌患者微小核糖核酸(miR)-34a、miR-145、miR-21及Ki-67的表达与人乳头瘤病毒(HPV)感染的关系.方法选择2020年1月-2022年11月新乡市中心医院收治的189例宫颈癌患者纳入宫颈癌组,宫颈上皮内瘤变(CIN)患者190例纳入CIN组,宫颈炎患者195例纳入宫颈炎组,检测三组患者miR-34a、miR-145、miR-21及Ki-67表达情况,分析宫颈不同病变HPV阳性率及HPV-DNA负荷量表达.结果宫颈炎组、CIN组、宫颈癌组miR-34a、miR-145表达水平呈降低趋势,miR-21表达水平及Ki-67阳性率呈升高趋势(P<0.05);随着病情加重,HPV阳性率和HPV-DNA表达水平逐渐升高(P<0.05);HPV阳性宫颈癌患者miR-34a、miR-145表达水平低于HPV阴性患者,miR-21表达水平高于HPV阴性患者(P<0.05).结论miR-34a、miR-145、miR-21及Ki-67的表达及HPV感染可参与慢性子宫颈炎-CIN-宫颈癌的病情发展,且miR-34a、miR-145、miR-21表达水平与HPV感染有关,可作为临床诊治的新靶点. OBJECTIVE To observe the expressions of micro ribonucleic acid-34a(miR-34a),miR-145,miR-21 and Ki-67 in the patients with cervical cancer and explore the association with human papillomavirus(HPV)infection.METHODS Totally 189 cervical cancer patients who were treated in Xinxiang Central Hospital from Jan 2020 to Nov 2022 were assigned as the cervical cancer group,190 patients who had cervical intraepithelial neoplasias(CIN)were assigned as the CIN group,and 195 patients with cervicitis were chosen as the cervicitis group.The expression levels of miR-34a,miR-145,miR-21 and Ki-67 were detected for the three groups of patients,and the positive rate of HPV and HPV-DNA load were observed and compared among the three groups.RESULTS The expression levels of miR-34a and miR-145 of the cervicitis group,the CIN group and the cervical cancer group showed downward trends,while the expression level of miR-2l and the positive rate of Ki-67 showed upward trends(P<0.05).The positive rate of HPV and expression level of HPV-DNA were elevated with the aggravation of illness condition(P<0.05).The expression levels of miR-34a and miR-145 of the HPV-positive cervical cancer patients were lower than those of the HPV-negative patients,while the expression level of miR-21 of the HPV-positive patients was higher than that of the HPV-negative patients(P<0.05).CONCLUSION The expressions of miR-34a,miR-145,miR-21 and Ki-67 as well as the HPV infection may participate in the progression of chronic cervicitis-CIN-cervical cancer;the expression levels of miR-34a,miR-145 and miR-21 are associated with the HPV infection,which can be served as the new targets for clinical diagnosis and treatment.
