miR-21/PTEN/Akt信号通路水平与Hp感染结石性胆囊炎胆囊黏膜病变的关系

Association between miR-21/PTEN/Akt signaling pathways and Hp infection-induced gallbladder mucosal lesions of calculous cholecystitis

摘要 目的基于miR-21/PTEN/Akt信号通路探究幽门螺杆菌(Hp)感染对结石性胆囊炎胆囊黏膜病变的影响.方法选择2017年1月-2020年1月中国医科大学附属盛京医院收治的结石性胆囊炎患者181例,根据胆囊组织是否感染Hp分为Hp感染组和Hp未感染组,比较两组患者胆囊黏膜病变情况,检测胆囊黏膜磷脂酶A2(PLA2)活性、胆囊壁收缩素-A(CCK-A)受体表达、胆囊排空率和微小RNA(miRNA)-21/磷酸酶与张力蛋白同源物基因(PTEN)/Akt信号通路表达情况.结果Hp感染组上皮化生阳性率高于Hp未感染组(P<0.05);Hp感染组胆囊黏膜PLA2活性高于Hp未感染组(P<0.05),胆囊壁CCK-A受体mRNA表达及胆囊排空率均低于Hp未感染组(P<0.05);Hp感染组胆囊黏膜miR-21、Akt mRNA相对表达量高于Hp未感染组(P<0.05),PTEN mRNA相对表达量低于Hp未感染组(P<0.05).结论Hp感染与结石性胆囊炎密切相关,可促进患者胆囊黏膜病变,其机制可能与miR-21/PTEN/Akt信号通路激活有关. OBJECTTVE To explore the influence of Helicobacter pylori(Hp)infectionon gallbladder mucosallesions of the patients with calculous cholecystitis based on miR-21/PTEN/Akt signaling pathway.METHODS A total of 18l patients with calculous cholecystitis who were treated in Shengjing Hospital of China Medical University from Jan 2017 to Jan 2020 were enrolled in the study and were divided into the Hp infection group and the no Hp infection group according to the status of Hp infection of gallbladder tissue.The status of gallbladder mucosal lesions was compared between the two groups of patients,the activity of phospholipase A2(PLA2)in gallbladder mucosa,expression level of cholecystokinin-A(CCK-A)receptor in gallbladder wall,gallbladder emptying rate and expression levels of microRNA(miRNA)-21/phosphatase and tensin hmmlogydeleted on ten(PTEN)/Akt signaling pathways were detected.RESULTS The positive rate of gallbladder metaplasia of the Hp infection group was higher than that of the no Hp infection group(P<0.05).The activity of gallbladder mucosa PLA2 of the Hp infection group was higher than that of the no Hp infection group(P<0.05).The expression level of CCK-A receptor mRNA in gallbladder wall and gallbladder emptying rate were lower in the Hp infection group than in the no Hp infection group(P<0.05).The relative expression levels of miR-21 and Akt mRNA in gallbladder mucosa were higher in the Hp infection group than in the no Hp infection group(P<0.05);the relative expression level of PTEN mRNA of the Hp infection group was lower than that of the no Hp infection group(P<0.05).CONCLUSION The Hp infection is closely associated with the calculous cholecystitis,it can accelerate the lesions of gallbladder mucosa,and the mechanisms may be associated with the activation of miR-21/PTEN/Akt signaling pathways.
