摘要
目的 观测当归补血汤(Danggui Buxue Decoction, DBD)口服对D-半乳糖致衰老小鼠皮肤抗氧化酶SOD、POD、CAT活性变化,探究DBD抗衰老的作用机制。方法 将小鼠40只造模成功后随机编号分为4组,即对照组、DBD低剂量组、DBD中剂量组、DBD高剂量组,每组各10只,对照组于同一时间点等量生理盐水灌胃;DBD低剂量组、DBD中剂量组、DBD高剂量组分别予DBD水煎剂生理盐水稀释1:1、2:1、3:1口服,观察4组小鼠皮肤表皮厚度、真皮厚度和胶原纤维总面积及小鼠皮肤抗氧化酶SOD、POD、CAT活性与mRNA相对表达量及与DBD水煎剂浓度相关性。结果 与实验前比较,实验第21天小鼠皮肤表皮厚度、真皮厚度和胶原纤维总面积差异有统计学意义(P<0.05);小鼠皮肤抗氧化酶SOD、POD、CAT活性差异有统计学意义(P<0.05);SOD、POD、CAT mRNA表达量差异有统计学意义(P<0.05)。结论 DBD可通过提高皮肤抗氧化酶SOD、POD、CAT活性,延缓皮肤退行性变化,从而延缓衰老进程。
Objective This paper aims to observe the effect of oral Danggui Buxue Decoction(DBD) on the activities of superoxide dismutase(SOD), peroxidase dismutase(POD) and catalase(CAT) in skin of D-galactose-induced aging mice, and explore the anti-aging mechanism of DBD. Methods Forty mice were randomly divided into 4 groups, including control group, low DBD dose group, medium DBD dose group and high DBD dose group, with 10 mice in each group. The control group was given the same amount of saline at the same time point. low DBD dose group, medium DBD dose group and high DBD dose group received DBD diluted with 1:1 saline, 2:1 saline and 3:1 saline, respectively. The epidermal thickness, dermal thickness and total area of collagen fibers of the 4 groups of mice were observed, and the correlation between the activities of antioxidant enzymes SOD, POD and CAT and the relative expression of mRNA and the concentration of DBD decoction in the skin of mice was observed. Results On the 21^(st) day of the experiment, there was statistical significance on the difference between epidermal thickness, dermal thickness and total area of collagen fibers of mouse skin compared with pre-experiment(P<0.05). There was statistical significance on the difference of SOD, POD and CAT activities in skin of mice(P<0.05). There was statistical significance on the difference of mRNA expression of SOD, POD and CAT(P<0.05). Conclusion DBD may delay skin degeneration by improving the activities of superoxide dismutase, peroxidase dismutase and catalase, so as to delay the aging process.
