摘要
目的:探讨壮药白苞蒿水提液及含药血清对小鼠离体子宫平滑肌收缩功能的影响,并初步探讨其收缩子宫平滑肌的作用机制。方法:利用雌二醇增加子宫的敏感度,采用生物采集系统记录小鼠离体子宫平滑肌的收缩曲线,观察壮药白苞蒿水提液及其血清对小鼠离体子宫平滑肌收缩频率、收缩幅度、收缩面积及活动力的影响;观察壮药白苞蒿水提液对肾上腺素干预下小鼠离体子宫平滑肌收缩频率、收缩幅度及活动力的影响。结果:(1)与同组给药前自身相比,空白对照组所有检测指标均无显著差异(P>0.05);白苞蒿15.00、7.50、3.75 mg/mL组可明显升高子宫平滑肌最大收缩张力、平均收缩张力、收缩面积、收缩频率、活动力、平均收缩张力促进率及子宫活力促进率(P<0.05或P<0.01),15.00 mg/mL组可明显升高最小收缩张力(P<0.05),3.75 mg/mL组可显著提高子宫平滑肌的活动力(P<0.05),3.75、7.50、15.00 mg/mL组对小鼠离体子宫活力促进率分别为:50.10%、122.60%、675.10%,平均收缩张力促进率分别为:17.60%、43.40%、129.30%。(2)AD 0.0025 mg/mL组的最大收缩张力和平均收缩张力均显著降低(P<0.01);与AD 0.0025 mg/mL组对比,白苞蒿25.00 mg/mL组可显著提高子宫平滑肌的最大收缩张力和平均收缩张力(P<0.01)。(3)与同组给药前相比,加入AD后,各组所有检测指标较造模前均明显降低(P<0.05或P<0.01);与同组造模后相比,空白对照组所有检测指标均无显著变化(P>0.05),白苞蒿15.00、7.50 mg/mL组可显著升高最大收缩张力、平均收缩张力、收缩频率和活动力(P<0.05或P<0.01)。(4)与同剂量空白血清相比,白苞蒿20 g/kg的含药血清50μL、100μL、200μL组对增强小鼠离体子宫的最大收缩张力优于空白血清(P<0.05),200μL组对增强小鼠离体子宫平均收缩张力优于空白血清(P<0.05),100μL组对小鼠离体子宫收缩频率和活动力优于空白血清(P<0.05)。结论:壮药白苞蒿及其含药血清均能兴奋小鼠离体子宫平滑肌,促进子宫收缩,有拟缩宫素样作用。
Objectives:To explore the effects of the aqueous extract and medicated serum of Artemisia lactiflora on the contractile function of isolated uterine smooth muscle in mice(IUSMM)and to preliminarily investigate the mechanism.Methods:Estrogen was used to increase the sensitivity of uterus.The contraction curve of IUSMM was recorded by the BL-420 biological acquisition system.The effects of the aqueous extract and medicated serum of A.lactiflora on the contraction frequency,contraction amplitude,contraction area,and activity of IUSMM were observed,and the effect of the aqueous extract on the contraction frequency,contraction amplitude and activity of IUSMM intervened by epinephrine was also explored.Results:After treatment,there were no significant differences in all the detection indexes in the blank control group(P>0.05),while 15,7.5 and 3.75 mg/mL A.lactiflora had an increase in the maximum contraction tension,mean contraction tension,contraction area,contraction frequency and activity as well as an elevation in the rates of promoting mean contraction tension and activity(P<0.01 or P<0.05);15 mg/mL A.lactiflora increased the minimum contraction tension(P<0.05),and 3.75 mg/mL A.lactiflora enhanced the activity of uterine smooth muscle(P<0.05).The rates of promoting the isolated uterine activity by 3.75,7.5 and 15 mg/mL A.lactiflora were 50.10%,122.60%and 675.10%,respectively,and those of mean contraction tension were 17.60%,43.40%and 129.30%,respectively.The maximum and mean contraction tensions in 0.0025 mg/mL A.lactiflora group were reduced(P<0.01).The 25 mg/mL A.lactiflora group had higher maximum and mean contraction tensions of uterine smooth muscle than the 0.0025 mg/mL A.lactiflora group(P<0.01).After treatment with A.lactiflora,all the detection indexes in each group were lower than those before modeling(P<0.05 or P<0.01).Compared with the conditions after modeling,there was no significant changes in all the detection indexes in the blank control group(P>0.05),while 15 and 7.5 mg/mL A.lactiflora elevated the maximum contraction tension,mean contraction tension,contraction frequency and activity(P<0.05 or P<0.01).Compared with the blank serum of the same dose,200μL,100μL and 50μL medicated serum showed better performance in enhancing the maximum contraction tension of mouse isolated uterus(P<0.05);200μL medicated serum group had better performance in promoting mean contraction tension(P<0.05);100μL medicated serum had better performance in promoting contraction frequency and activity(P<0.05).Conclusion:A.lactiflora and its medicated serum can activate the IUSMM and promote uterine contraction,showing oxytocin-like effect,which facilitate the treatment of functional hemorrhage.
