波罗蜜籽果胶提取工艺优化及抗氧化活性研究

Optimization of Extraction Technology and Antioxidant Activity of Pectin from Jackfruit Seed

摘要 为发掘波罗蜜籽的工业应用价值,采用超声波辅助酸液法提取波罗蜜籽中的果胶,并考察果胶对亚硝酸根离子(NO_(2)^(-))、羟基自由基(·OH)的清除能力和对油脂酸败反应的抑制效果以衡量其抗氧化活性。依据单因素试验结果,采用Box-Behnken法优化提取工艺,得到最佳提取工艺参数:p H3、料液比1:36 g/mL、提取温度46℃、超声时间16 min、超声功率200 W,在此条件下波罗蜜籽果胶平均提取率为13.84%,与模型预测值14.73%相近。波罗蜜籽果胶的抗氧化活性与其浓度成正相关。当波罗蜜籽果胶浓度为2.2 mg/mL时,对NO_(2)^(-)和·OH的清除率分别为58%、88%。波罗蜜籽果胶可推迟油脂酸败反应的诱发期,延长油脂在自然环境中的保存时间。研究结果表明,波罗蜜籽果胶提取率良好,且具有良好的抗氧化活性。 In order to explore the industrial application value of jackfruit seeds,the ultrasonic assisted acid solution method was used to extract pectin from jackfruit seeds,and the scavenging ability of pectin to nitrous acid ions,hydroxyl radicals and the inhibition effect on oil rancidity were investigated to measure its antioxidant activity.According to the results of single factor experiment,and the optimal extraction process parameters which used by Box Behnken method were obtained:pH3,material liquid ratio 1:36 g/mL,extraction temperature 46℃,ultrasonic time 16 min,and ultrasonic power 200 W.Under this condition,the average extraction rate of pectin from jackfruit seeds was 13.84%,close to the model prediction value of 14.73%.The antioxidant activity of pectin from jackfruit seeds is positive correlation to its concentration.When the concentration of pectin in jackfruit seed was 2.2 mg/mL,the clearance rates of NO_(2)^(-) and·OH were 58% and 88% respectively.The pectin from jackfruit seed can delay the induction period of rancidity reaction of oil and prolong the storage time of oil in the natural environment.The research results showed that the extraction rate of pectin from jackfruit seeds was good,and it had good antioxidant activity.
