妊娠期高血压对子代大鼠肾组织端粒长度的影响

摘要 目的探讨妊娠期高血压对子代大鼠肾组织端粒长度的影响。方法将6只8周龄SD雌性大鼠随机等分为实验组和对照组,实验组大鼠构建高血压模型。分别于术前及术后1个月测量两组雌鼠的鼠尾血压。术后1个月时,选取3只10周龄雄鼠,每只雄鼠均随机与对照组中的1只雌鼠和实验组中的1只雌鼠合笼饲养,待雌鼠生产后,从每只雌鼠的子代中随机选取4只新生大鼠,构成子代实验组和子代对照组,采用实时荧光定量PCR法检测子代大鼠肾组织端粒的相对长度、端粒酶及端粒结合蛋白的相对表达量。结果术后1个月,实验组大鼠血压高于对照组,差异有统计学意义(t=5.266,P<0.05)。实验组与对照组雌鼠子代数量分别为(11.33±2.15)只和(12.67±3.10)只,差异无统计学意义(t=0.859,P>0.05)。与子代对照组比较,子代实验组大鼠肾组织端粒相对长度较短,差异有统计学意义(0.82±0.09 vs.1.04±0.07,t=3.024,P<0.05),肾组织端粒酶逆转录酶相对表达量较高,差异有统计学意义(2.11±0.58 vs.1.01±0.34,t=7.248,P<0.05)。与子代对照组比较,子代实验组端粒重复因子1(TRF1)、端粒重复因子2(TRF2)、端粒保护因子1(POT1)、端粒保护蛋白1(TPP1)表达水平均升高,差异均有统计学意义(均P<0.05),两组TRF1相互作用核蛋白2(TIN2)和阻滞活化蛋白1(RAP1)表达水平比较差异均无统计学意义(均P>0.05)。结论亲代雌鼠妊娠期高血压可造成子代大鼠肾组织端粒长度缩短,对肾脏发育造成潜在影响。 Objective To explore the ffct of hypertension during pregnancy on telomere length of renal tisue in offspring rats.Methods Six 8-week-old female SD rats were randomly divided into experimental goup and control group.Hypertension model was built in experimental group.The tail blood pressure levels of female rals in the two groups were measured before operation and al one month after operation.Al one moonth after operation,three 10-week-old male rats were selected,and each male rat was randomly caged with one fe-male rat in control group and one female rot in experinental group.After delivey.four newborn rals were rundomly seleeted frorm the of_springs of each female ratls t0 form the ofspring experimental group and the ofspring control group.Real-time fluorescence quantitative PCR was used to deet the relative length of telomere,the relative epession of elomerase and telomere binding prolein in renal tsss of the ofF spring rats.Results Al one month alfer operation,blood pressure of female rats in experimental group was signifcanly higher than that in control group(t=5.266,P<0.05).The number of offspring rats in experimental group and control group were(11.33+2.15)and(12.67+3.10),respectively,there was no significant dfference(1=0859,P>0.05).Compared with olfspring control grup,the relative lengh of telomere in renal isse of ofspring experimental goup was shorter(0.82+0.098.1.04+0.07,1=3.024,P<0.05),the expres-siom of telomerase reverse transcripase was higher(21120.58 ts.1.01+0.34.1=7.248.P<0.05).Compured with fepring control goup,the epesion levels of telomere repeatl factor 1(TRF1),tlonere repeat factor2(TRF2),telonere protetive factor1(POTI).nd Teluumurc procctirc protcin 1(TPP1)in ofopring erpcrimcntal group inoreaoed ognificuinly(all Px0.05),there vra no aignificant diferenee in expression levels of TRF1 interacting mucleoprotein2(TIN2)and retarded activation protein1(RAPI)between the two groups(all P>0.05).Conclusion Hypertension during pregnaney can shorten telonere length of renal tssue in ofsping rats,which has poten-tial impact on renal development.
