摘要
[目的]探讨蜂毒素对热应激鹌鹑睾丸抗氧化功能、热休克蛋白和紧密连接的影响,为蜂毒素在热应激鹌鹑中的应用及效果提供科学依据。[方法]选取120只20日龄雄性鹌鹑,随机分为3组,每组4个重复,每个重复10只鹌鹑,对照组(Con组)的环境温度为(24±2)℃,热应激组(HS组)和热应激+蜂毒素组(HSM组)每天12:00-16:00进行4 h热应激,其温度为(36±2)℃,其余时间温度为(24±2)℃,持续10 d。[结果]HS组与Con组相比,热应激显著降低鹌鹑睾丸的重量、器官指数、SOD活性、ZO-1和Occludin mRNA相对表达水平,显著增加MDA浓度和HSP70和HSP90 mRNA相对表达量;HSM组与HS组相比,饲粮中添加蜂毒素能显著增加热应激鹌鹑睾丸的重量、器官指数、GSH-Px活性、ZO-1 mRNA相对表达水平,显著降低MDA活性、HSP70和HSP9 mRNA相对表达量。[结论]日粮中添加蜂毒素可以改善热应激对鹌鹑睾丸器官指数、抗氧化功能和紧密连接的不利影响,从而提高鹌鹑在热应激下的繁殖性能。
[Objectives]This article aimed at studying the effects of melittin on the antioxidant function,heat shock protein and tight junction in heat stressed quail testes to provide scientific basis for the application and effectiveness of melittin in heat stressed quails.[Methods]120 male quails of 20 day old were randomly divided into 3 groups,with 4 replicates in each group and 10 in each replicate.The ambient temperature of the control group(Con group)was(24±2)℃.The heat stress group(HS group)and the heat stress+melittin group(HSM group)were subjected to 4 hours of heat stress with a temperature of(36±2)℃ from 12:00 to 16:00 every day and a temperature of(24±2)℃ for the rest of the time,lasting for 10 days.[Results]Heat stress significantly reduced the weight,organ index,SOD activity,relative expression levels of and mRNA,and significantly increased MDA concentration and relative expression levels of HSP70 and HSP90 mRNA in the quail testes in the HS group compared with the Con group.Adding melittin to the diet significantly increased the weight,organ index,GSH-Px activity,and relative expression level of ZO-1 mRNA,while significantly reducing MDA activity and relative expression levels of HSP70 and HSP90 mRNA in the heat stressed quail testes in the HSM group compared with the HS group.[Conclusions]Adding melittin to the diet improved the adverse effects of heat stress on the organ index,antioxidant function,and tight junction of quail testes,thereby improving their reproductive performance under heat stress.
