非编码RNA在胃肠肿瘤微环境中免疫细胞的作用机制研究

摘要 目的观察长链非编码RNA(long-non-coding RNA,lncRNA)在胃肠肿瘤微环境中免疫细胞的作用机制,明确lncRNA以及同源基因HOXA远端转录本(HOXAtranscript at the distal tip,HOTTIP)、甘油醛-3磷酸脱氢酶(glycaraldehyde-3Phosphate dehydrogenase,GAPDH)在胃肠肿瘤中的表达。方法本研究为大鼠试验,于2022年4—12月选择100只成年雄性大鼠作为研究对象,长期喂养高硝酸盐食物,形成胃肠肿瘤后,进行胃癌手术或者结肠癌手术,探讨非编码RNA(non-coding,ncRNA)在胃肠肿瘤细胞中的表达并与癌旁组织表达能力对比,基于表达能力的不同,对比胃肠肿瘤大鼠的生存时间,最后对比RUNX1-IT1与Lv-NC细胞迁移增殖能力。结果lncRNA及同源基因在胃肠肿瘤中的表达结果中,HOTTIP在胃癌组织中的表达为(3.99±1.05),在癌旁组织的表达为(3.50±1.03),差异有统计学意义(t=2.356,P=0.021)。HOTTIP与GAPDH的引物都表现为单峰,有着较高的特异性;lncRNA及其同源基因在胃肠肿瘤预后中,使用Kaplan-Meier对预后与基因表达关系进行的研究中总生存率比较,差异无统计学意义(P>0.05);在迁移实验及增殖实验中Lv-RUNX1-IT1细胞的迁移能力和增殖能力都有所降低。结论非编码RNA在胃肠肿瘤微环境当中,可有效抑制胃肠肿瘤细胞表达,对大鼠生存时间未造成较大影响,Lv-RUNX1-IT1细胞的迁移能力和增殖能力有所降低。 Objective To observe the mechanism of long-non-coding RNA(lncRNA)in gastrointestinal tumor microenvironment of immune cells,and to clarify the expression of lncRNA as well as the homologous genes HOXAtranscript at the distal tip(HOTTIP)and glycaraldehyde-3Phosphate dehydrogenase(GAPDH)in gastrointestinal tumors.Methods This study was a rat test,and selected 100 adult male rats from April to December 2022 as the study subjects.After feeding high nitrate food for a long time and forming gastrointestinal tumors,gastric cancer surgery or colon cancer surgery was performed.We explored the expression of non-coding RNA(ncRNA)in gastrointestinal tumor cells,and compared the expression ability with that of paracancerous tissues,and based on the difference in expression ability,we compared the survival time of rats with gastrointestinal tumors,and finally,we compared the migratory and value-added ability of RUNX1-IT1 with that of Lv-NC cells.Results Among the expression results of LncRNA and homologous genes in gastrointestinal tumors,the expression of HOTTIP was(3.99±1.05)in gastric cancer tissues and(3.50±1.03)in paracancerous tissues,the difference was statistically significant(t=2.356,P=0.021).The primers for both HOTTIP and GAPDH showed single peaks with high specificity.In the prognosis of gastrointestinal tumors,there was no statistically significant difference in the overall survival rate between lncRNA and its homologous genes in the study of the relationship between prognosis and gene expression using Kaplan-Meier(P>0.05).The migration ability and proliferation ability of Lv-RUNX1-IT1 cells were decreased in migration and proliferation experiments.Conclusion Non-coding RNAs can effectively inhibit the expression of gastrointestinal tumor cells in the microenvironment of gastrointestinal tumors,which did not have a large impact on the survival time of rats,and the migration and value-added ability of Lv-RUNX1-IT1 cells were reduced.
