基于生物信息学分析幽门螺杆菌感染与骨质疏松症的相关性

摘要 目的 利用生物信息学分析幽门螺杆菌(HP)感染与骨质疏松症(OP)的关系。方法 通过Disgenet、OMIM、TTD、GeneCards、Pharmgkb等基因数据库查找引起OP和HP感染的疾病基因,构建疾病共同基因的蛋白互作网络并筛选出关键靶点基因,通过DAVID数据库进行基因本体论(GO)功能和京东基因与基因组百科全书(KEGG)通路富集分析。结果 通过相关基因数据库筛选出获得OP相关基因493个,HP感染相关基因798个,通过取交集后获得69个HP感染与OP共同靶点基因。GO功能主要富集在序列特异性DNA结合转录因子活性的正向调节、Ⅰ-κB激酶/核因子κB信号传导等方面,KEGG通路包括人巨细胞病毒感染、沙门菌感染等信号通路。HP感染与OP的关键靶点基因为丝氨酸/苏氨酸蛋白激酶1、清蛋白、CTNNB1、雌激素受体1、白细胞介素1B、白细胞介素6、瘦素、基质金属蛋白酶-9、MYC、肿瘤坏死因子等。结论 HP感染多通过导致炎性反应产生炎症因子,从而作用于影响骨吸收及骨形成的信号通路,最终导致或加重OP。 Objective To analyze the relationship between Helicobacter pylori(HP)infection and osteoporosis(OP)by bioinformatics.Methods Disease genes causing OP and HP infection were searched through gene databases such as Disgenet,OMIM,TTD,GeneCards,Pharmgkb and other gene databases.The protein interaction networks of common disease genes were constructed and key target genes were screened.Gene ontology(GO)function and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed through the DAVID database.Results A total of 493 OP-related genes and 798 HP infection-related genes were screened out through the related gene database,and 69 common target genes of HP infection and OP were obtained through intersection.GO function was mainly concentrated in the positive regulation of sequence-specific DNA-binding transcription factor activity and the signaling ofⅠ-κB kinase/nuclear factor-κB.KEGG pathway includes human cytomegalovirus infection and salmonella infection.The key target genes of HP infection and OP were serine/threonine protein kinase 1,albumin,CTNNB1,estrogen receptor 1,interleukin-1B,interleukin-6,leptin,matrix metalloproteinase-9,MYC,tumor necrosis factor,etc.Conclusion HP infection usually produces inflammatory factors by leading to inflammatory response,thus acting on the signaling pathways affecting bone resorption and bone formation,and ultimately causing or aggravating OP.
