miRNA-494-3p靶基因预测及其相关信号通路的生物信息分析

Bioinformatic Analysis of Target Gene Prediction and Related Signaling Pathways of miRNA-494-3p

摘要 目的采用生物信息学的方法对miRNA-494-3p的靶基因进行预测,并对靶基因集合进行功能富集分析和信号通路分析。方法应用微小RNA(miRNA)靶基因数据库StarBase进行靶基因预测,选取miRanda、PITA和Targetscan 3个数据库预测结果的交集作为进一步分析的基因集合。利用DAVID数据库,对靶基因进行基因本体(GO)分析和京都基因与基因组百科全书(KEGG)信号通路富集分析。结果3个数据库预测到miRNA-494-3p的靶基因253个,其功能分别富集在蛋白质结合、双链DNA结合、RNA结合等分子功能,RNA聚合酶Ⅱ的转录调控、细胞增殖的正调控、细胞分化、细胞凋亡等生物学过程以及细胞核、核质、胞浆、细胞质、细胞隔膜等细胞组分上;生物学通路主要富集于癌症通路、FoxO信号通路、细胞吞噬作用、刺猬信号通路、细胞黏附连接、肌动蛋白细胞骨架的调控、谷氨酸能突触、昼夜节律以及p53信号通路中。结论miRNA-494-3p的功能主要富集在与癌症、胰岛素抵抗、糖尿病、炎症、胚胎发育等相关的多个信号通路中,为进一步研究其对靶基因和信号通路的调控作用及其可能的机制提供了线索。 Objective Bioinformatics methods were used to predict the target genes of mirna-494-3p,and functional enrichment analysis and signal pathway analysis were performed on the target gene set.Methods The miRNA target gene database Starbase was used for target gene prediction,and the intersection of the prediction results of miRanda,PITA and Targetscan was selected as the gene set for further analysis.Using David database,Gene Ontology analysis and KEGG signal transduction pathway enrichment analysis were performed on target genes.Results The three databases predicted 253 target genes of mirna-494-3p.Their functions were enriched in protein binding,double stranded DNA binding,RNA binding and other molecular functions;transcriptional regulation of RNA polymerase I,positive regulation of cell proliferation,cell differentiation,apoptosis and other biological processes;as well as cell components such as nucleus,nucleoplasm,cytoplasm,cytoplasm and cell membrane.Biological pathways are significantly enriched in cancer pathway,FOXO signaling pathway,phagocytosis,hedgehog signaling pathway,cell adhesion and connection,regulation of actin cytoskeleton,glutamatergic synapse,circadian rhythm,and p53 signaling pathway.Conclusion The function of miRNA-494-3p is mainly concentrated in multiple signaling pathways related to.cancer,insulin resistance,diabetes,inflammation,and embryonic development,providing clues for further study of its regulatory effects on target genes and signaling pathways and its possible mechanisms.
