摘要
目的:探讨溃疡性结肠炎(Ulcerative colitis,UC)患者结肠黏膜C-C基序趋化因子配体2(C-C motif chemokine ligand 2,CCL-2)、白细胞介素-23(Interleukin-23,IL-23)、基质Gla蛋白(Matrix Gla protein,MGP)表达与临床参数的关系。方法:收集2018年4月至2022年10月医院收治的103例UC患者结肠黏膜组织记为研究组,同一时期单发息肉切除后行结肠镜复查的58例健康者的正常黏膜组织记为对照组。采用实时荧光定量聚合酶链式反应(Real time fluorescence quantitative Polymerase chain reaction,RT-PCR)检测结肠黏膜组织CCL-2、IL-23及MGP mRNA表达水平,分析其与UC患者病情分期、病变范围及内镜分级的关系。结果:研究组结肠黏膜组织CCL-2、IL-23及MGP mRNA表达水平高于对照组(P<0.05),且与UC患者病情分期呈正相关(r=0.732、0.748、0.739,P=0.000、0.000、0.000),与UC患者病变范围呈正相关(r=0.515、0.401、0.389,P=0.000、0.000、0.000),与UC患者内镜分级呈正相关(r=0.681、0.623、0.602,P=0.000、0.000、0.000)。结论:UC患者结肠黏膜CCL-2、IL-23、MGP表达与病情分期、病变范围及内镜分级有关。
Objective:To investigate the relationship between the expression of C-C motif chemokine ligand 2(CCL-2),interleukin-23(IL-23),matrix Gla protein(MGP),and clinical parameters in patients with Ulcerative colitis(UC).Methods:A total of 103 UC patients admitted to our hospital from April 2018 to October 2022 were collected as the study group,while 58 healthy individuals who underwent colonoscopy after single polypectomy during the same period were included as the control group.Real-time fluorescence quantitative Polymerase chain reaction(RT-PCR)was used to detect the expression levels of CCL-2,IL-23,and MGP mRNA in the colonic mucosa of the two groups.The relationship between the expression levels of CCL-2,IL-23,and MGP in colonic mucosal tissue and the stage,range,and endoscopic grading of UC patients were analyzed.Results:The mRNA expression levels of CCL-2,IL-23,and MGP in the colon mucosa tissue of the study group were higher than those of the control group(P<0.05)and were positively correlated with the disease stage of UC patients(r=0.732,0.748,0.739,P=0.000,0.000,0.000),and positively correlated with the extent of lesions in UC patients(r=0.515,0.401,0.389,P=0.000,0.000,0.000),and positively correlated with endoscopic grading in UC patients(r=0.681,0.623,0.602,P=0.000,0.000,0.000).Conclusion:The expression of CCL-2,IL-23,and MGP in colon mucosa of UC patients is related to disease stage,lesion range,and endoscopic grading.
