胃癌组织MLH1、MSH2、MSH6和PMS2蛋白表达意义及与患者病理特征的关系研究

Expression of MLH1,MSH2,MSH6 and PMS2 proteins in gastric cancer and their relationship with pathological characteristics

目的探讨胃癌组织4种错配修复蛋白(MLH1、MSH2、MSH6和PMS2)表达意义及与患者病理特征的关系。方法选择2021年6月至2022年6月浙江省舟山医院收治的101例胃癌患者临床病理资料,收集经手术切除的胃癌组织及配对癌旁组织病理标本。采用免疫组织化学法检测并比较癌组织与癌旁组织标本MLH1、MSH2、MSH6和PMS2蛋白阳性率;分析不同病理特征胃癌患者之间MLH1、MSH2、MSH6和PMS2蛋白阳性率的差异。结果胃癌组织MLH1、MSH2、MSH6和PMS2蛋白阳性率均高于癌旁组织(均P<0.05)。不同性别、年龄、肿瘤直径和分化程度胃癌患者组织MLH1、MSH2、MSH6和PMS2蛋白阳性率比较,差异均无统计学意义(均P>0.05);Ⅲ~Ⅳ期胃癌患者组织MLH1、MSH2、MSH6和PMS2蛋白阳性率高于Ⅰ~Ⅱ期患者,淋巴结转移胃癌患者组织MLH1、MSH2、MSH6和PMS2蛋白阳性率高于无淋巴结转移患者,浸润至肌层/浆膜层胃癌者组织MLH1、MSH2、MSH6和PMS2蛋白阳性率高于黏膜/黏膜下层患者,差异均有统计学意义(均P<0.05)。结论胃癌组织MLH1、MSH2、MSH6和PMS2蛋白高表达,且与临床分期、淋巴结转移和浸润深度有关。

Objective To investigate the expression of mismatch repair proteins MLH1,MSH2,MSH6 and PMS2 in gastric cancer and their relationship with pathological characteristics.Methods Clinical and pathological data of 101 gastric cancer patients admitted to Zhejiang Zhoushan Hospital from June 2021 to June 2022 were retrospectively reviewed.The expression of MLH1,MSH2,MSH6 and PMS2 proteins in cancer tissue and adjacent tissue samples was detected with immunohistochemical method and compared between two groups;the correlation of MLH1,MSH2,MSH6 and PMS2 expression with pathological characteristics of gastric cancer was analyzed.Results The positive rates of MLH1,MSH2,MSH6 and PMS2 proteins in gastric cancer tissues were significantly higher than those in adjacent tissues(all P<0.05).There were no significant differences in the positive rates of MLH1,MSH2,MSH6 and PMS2 proteins among patients with different genders,ages,tumor diameters and degrees of differentiation(all P>0.05);while the positive rates in stageⅢ-Ⅳpatients were higher than those in stageⅠ-Ⅱpatients;in patients with lymph node metastasis were higher than those without lymph node metastasis;in patients with muscle/serous layer invasion were higher than those with mucosal/submucosal layer invasion(all P<0.05).Conclusion The expression of MLH1,MSH2,MSH6 and PMS2 protein is upregulated in gastric cancer,which is closely related to clinical stage,lymph node metastasis and invasion depth.
