摘要
为了解2种不同养殖环境下中华圆田螺肠道微生物多样性,设置稻田养殖组(A)池塘养殖组(B),别取A组和B组田螺的肠道内容物进行总菌群DNA提取纯化,采用高通量测序技术进行16S V3-V4区域扩增子测序以及生物信息学分析。结果表明,2组样本共得到131240条序列,共涉及25门215属,其中共有菌属169种,特有菌属各23种。基于97%的样本相似度,样本优化聚类为609个OTU(Operational taxonomical unit)。A组以拟杆菌门(51.32%)、变形菌门(32.15%)为主;B组以拟杆菌门(43.12%)、变形菌门(25.12%)、厚壁菌门(18.57%)为主。在属水平上,A组以拟杆菌类(Bacteroides)、气单胞菌(Aeromonas)、乳酸菌属(Cetobacterium)等属细菌为主,B组以拟杆菌类(Bacteroides)、气单胞菌(Aeromonas)、变形菌属(Proteocatella)等属细菌为主。指出,A组肠道菌群比B组肠道菌群具有更高的个体差异性、更丰富的物种多样性、更高的菌群丰度。
In order to understand the intestinal microbial diversity of Cipangopaludina chinensis under two different breeding environments,a paddy field culture group(A)and a pond culture group(B)were set up.The intestinal contents of group A and group B were extracted and purified for total flora DNA.High-throughput sequencing technology was used for 16 S V3—V4 region amplicon sequencing and bioinformatics analysis.The results showed that a total of 131240 sequences were obtained from the two groups of samples,involving 25 phyla and 215 genera,including 169 common genera and 23 endemic genera.Based on 97%of the sample similarity,the sample optimization clustering was 609 OTUs(Operational taxonomical units).Group A was dominated by Bacteroidetes(51.32%)and Proteobacteria(32.15%).Group B was dominated by Bacteroidetes(43.12%),Proteobacteria(25.12%)and Firmicutes(18.57%).At the genus level,group A was dominated by Bacteroides,Aeromonas,and Cetobacterium,while group B was dominated by Bacteroides and Aeromon.
