摘要
转基因玉米MON87419是孟山都远东有限公司研发的转化体,转入了dom和pat基因,能够耐受麦草畏和草铵膦两种除草剂。本研究以转基因耐除草剂玉米MON874193′端特异性序列为靶标设计引物和探针,经特异性测试、体系优化、灵敏度测试及检出限(LOD)测试,建立了MON87419的实时荧光PCR定性检测方法。结果表明,该方法能检测出转基因耐除草剂玉米MON87419转化体成分,具有稳定性好、特异性强、灵敏度高的特点,检出限可达0.05%。该方法主要技术参数全部符合相关标准的要求,可为转基因玉米的安全监管提供有效的技术支撑。
Genetically Modified(GM)maize MON87419,developed by Monsanto Far East Co.,LTD.,which has been transformed into the dom and pat genes,is tolerant to two herbicides,wheat wort and glyphosate.In this study,primers and probes were designed based on the specific sequence of 3′end of GM herbieide-tolerant maize MON87419.Through specificity test,system optimization,sensitivity test and limit of detection(LOD)test,a qualitative detection method for MON87419 was established by real-time PCR.The results showed that the method could detect the components of GM herbicide-tolerant maize MON87419 with good stability,specificity and sensitivity.The LOD was 0.05%.The main technical parameters of this method all met the requirements of relevant standards,which could provide effective technical support for the safety supervision of GM maize.
