摘要
目的研究高脂饮食(high fat diet,HFD)雄性小鼠股骨骨密度、骨微结构及分形维数的改变。方法将3周龄雄性C57BL/6N小鼠按不同饮食分为正常饮食组(normal diet,ND)组和HFD组,在饲养过程中不同时间点采集样本。采血检测生化指标和糖代谢相关指标,对小鼠股骨进行骨小梁CT扫描,获得骨微结构及分形维数相关参数。结果与ND组比较,HFD组小鼠体质量(40.61±5.36 vs.54.69±2.28,P<0.05)、胆固醇(3.10±0.48 vs.5.33±0.80,P<0.05)、高密度脂蛋白(1.71±0.20 vs.2.15±0.14,P<0.05)和低密度脂蛋白(0.48±0.20 vs.1.10±0.21,P<0.05)在第24周后显著增加(P<0.05);股骨骨密度更低(0.15±0.02 vs.0.14±0.01,P<0.01)。骨微结构检测显示,HFD组小鼠股骨体积分数下降、骨小梁数量减少及厚度变薄,骨小梁分离度及模式结构指数增大,分形维数值下降。结论HFD雄性小鼠股骨骨量减少,骨微结构及FD下降。
Objective To study the changes of femoral bone mineral density,bone microstructure,and fractal dimension(FD)in male mice fed with high fat diet(HFD).Methods The 3-week-old male C57BL/6N mice were divided into normal diet(ND)group and HFD group according to different diets,and samples were collected at different time points in the feeding process.Blood biochemical indexes and related indexes of glucose metabolism were detected,and bone trabecular CT scanning was performed on the femur of mice to obtain related parameters of bone microstructure and FD.Results Compared with the ND group,the body mass(40.61±5.36 vs.54.69±2.28,P<0.05),cholesterol(3.10±0.48 vs.5.33±0.80,P<0.05),high-density lipoprotein(1.71±0.20 vs.2.15±0.14,P<0.05),and low-density lipoprotein(0.48±0.20 vs.1.10±0.21,P<0.05)of HFD group mice significantly increased after 24 weeks(P<0.05);The bone density of the femur is lower(0.15±0.02 vs.0.14±0.01,P<0.01).In the bone microstructure,the volume fraction of femur and the number of trabeculae decreased and the thickness became thinner in HFD group,suggesting that the bone mass decreased.The trabecular separation and structure model index(SMI)increased.The FD value decreased.Conclusion HFD cause the decrease of bone mass and impariment of bone microstructure in femurs of male mice.
