寄生原虫单细胞测序的研究进展

Research progress of single-cell sequencing technology in protozoon parasites

摘要 近年来随着单细胞测序技术迅速发展,其在人体、动物、植物、肿瘤及病原微生物等领域已经广泛开展研究。与宿主体内的基因突变相似,大多数寄生虫也存在突变,其基因组测序研究多集中在数百万大样本单个虫体细胞中。目前针对改变特定的宿主-寄生虫系统,现已开发出寄生虫单细胞测序方法,此举使得复杂的寄生虫种群中的遗传多样性和亲缘关系得以破译,并能捕获遗传突变的基因序列。本研究以人兽共患的疟原虫和利什曼原虫为例,概述寄生虫单细胞基因组测序的方法和进展,以及这些方法在寄生虫生物学中的应用。 In recent years,single-cell sequencing technology has developed rapidly,and studied widely in human,animal,plant,tumor and pathogenic microorganism.There are a large number of genetic mutations in the host,most parasites also have mutations,and their genome sequencing studies are focused on a large sample in millions of individual parasitic cells.Single-cell sequencing of parasites has been developed to modify specific host-parasite systems,which allows the genetic diversity and phylogenetic relationships in complex parasite populations to be deciphered and the genetic sequences of inherited mutations to be captured.Taking plasmodium and leishmania parasite as examples,this study summarizes the methods and progress of single-cell genome sequencing in protozoa parasites,and the application of these methods in parasitological biology.
