lncRNA TMPO-AS1在食管癌组织中的表达及临床意义

摘要 目的探讨长链非编码RNA(lncRNA)胸腺生成素反义RNA1(TMPO-AS1)在食管癌(EC)组织中的表达及临床意义。方法选取2016年8月至2019年8月南通大学附属启东医院收治的84例EC患者的EC组织和癌旁组织,检测EC组织和癌旁组织中TMPO-AS1、miR-498表达水平,分析EC组织中TMPO-AS1、miR-498表达水平与患者临床病理特征的关系,分析EC组织中TMPO-AS1与miR-498表达水平的相关性,分析EC组织中TMPOAS1、miR-498表达水平与患者预后的关系,分析影响EC患者预后的因素。结果与癌旁组织相比,EC组织中TMPO-AS1表达水平显著升高,miR-498表达水平显著降低(P均<0.05)。EC组织中TMPO-AS1、miR-498表达水平与肿瘤分化程度、TNM分期均有相关性(P均<0.05)。EC组织中TMPO-AS1与miR-498表达水平呈负相关(P<0.05)。TMPO-AS1高表达组、miR-498低表达组的累积生存率分别较TMPO-AS1低表达组、miR-498高表达组显著降低(P均<0.05)。TMPOAS1高表达、TNM分期Ⅲ期、miR-498低表达是影响EC患者预后的独立危险因素(P均<0.05)。结论EC组织中TMPO-AS1表达上调,miR-498表达下调,两者呈负相关。TMPO-AS1和miR-498有潜力成为EC患者病情评估、预测预后的辅助指标。 Objective This paper intends to explore the expression and clinical significance of long non-coding RNA(lncRNA)thymopoietin antisense RNA1(TMPO-AS1)in esophageal cancer(EC)tissue.Methods The EC tissue and paracancerous tissue of 84 EC patients admitted to Qidong Hospital Affiliated to Nantong University from August 2016 to August 2019 were selected.The expression levels of TMPO-AS1 and miR-498 in EC tissues and adjacent tissues were detected.The relationship between the expression levels of TMPO-AS1 and miR-498 in EC tissues and the clinical pathological characteristics of patients were analyzed.The correlation between the expression levels of TMPO-AS1 and miR-498 in EC tissues,and the relationship between the expression levels of TMPO-AS1 and miR-498 in EC tissues and patient prognosis were analyzed,so was the factors affecting the prognosis of EC patients.Results Compared with adjacent tissues,the expression level of TMPO-AS1 in EC tissues is significantly increased,while the expression level of miR-498 is significantly reduced(P<0.05).The expression levels of TMPO-AS1 and miR-498 in EC tissues are correlated with tumor differentiation and TNM staging(P<0.05).There is a negative correlation between the expression levels of TMPO-AS1 and miR-498 in EC tissues(P<0.05).The cumulative survival rates of the TMPO-AS1 high expression group and the miR-498 low expression group are significantly lower than those of the TMPO-AS1 low expression group and the miR-498 high expression group,respectively(P<0.05).The high expression of TMPO-AS1,TNM stage III,and low expression of miR-498 are independent risk factors affecting the prognosis of EC patients(P<0.05).Conclusions TMPO-AS1 expression is upregulated and miR-498 expression is downregulated in EC tissues,showing a negative correlation between the two.TMPO-AS1 and miR-498 have the potential to become auxiliary indicators for evaluating the condition and predicting prognosis of EC patients.
