环状RNA UBAP2在结直肠癌患者中的表达及与预后的关系

Expression of circular RNA UBAP2 in patients with colorectal carcinoma and its relationship with prognosis

目的探讨环状RNA泛素关联蛋白2(UBAP2)在结直肠癌患者中的表达及与预后的关系。方法以2018年6月至2019年6月在杨凌示范区医院住院接受手术的80例结直肠癌患者为研究对象,收集术前结直肠癌患者外周血标本、术中癌组织标本和癌旁组织标本。采用实时荧光定量聚合酶链反应(qRT-PCR)检测血清及结直肠癌组织、癌旁组织中环状RNA UBAP2表达水平,并分析癌组织中环状RNA UBAP2表达水平与患者临床特征的关系;对患者进行3年随访,采用Kaplan-Meier生存曲线分析结直肠癌组织中环状RNA UBAP2表达水平与患者预后的关系;采用多因素COX回归分析结直肠癌患者预后的影响因素。结果结直肠癌患者外周血中环状RNA UBAP2的表达水平(2.06±0.97)显著高于健康对照者(1.02±0.14),差异有统计学意义(P<0.05);与癌旁组织相比,结直肠癌组织中环状RNA UBAP2的表达水平显著升高(P<0.05);结直肠癌组织中环状RNA UBAP2的表达水平与TNM分期、淋巴结转移、浸润深度、分化程度有关(P<0.05)。Kaplan-Meier生存曲线分析显示,结直肠癌组织中环状RNA UBAP2高表达患者3年生存率低于低表达患者(P<0.05)。多因素COX回归分析显示,环状RNA UBAP2、TNM分期、淋巴结转移、浸润深度、分化程度是结直肠癌患者预后的影响因素(P<0.05)。结论结直肠癌患者外周血和癌组织中环状RNA UBAP2呈显著高表达,其中癌组织中环状RNA UBAP2表达水平与TNM分期、淋巴结转移、浸润深度、分化程度等临床病理特征及预后有密切联系,对判断患者预后情况具有重要价值。

Objective To investigate the expression of circular RNA ubiquitin-associated protein 2(UBAP2)in the patients with colorectal cancer and its relationship with prognosis.Methods A total of 80 inpatients with colorectal cancer undergoing hospitalization surgery in Yangling Demonstration Area Hospital from June 2018 to June 2019 served as the research subjects.The peripheral blood samples of the patients with colorectal cancer,intraoperative cancer tissue samples and paracancerous tissue samples were collected.The expression levels of circular RNA UBAP2 in serum,colorectal cancer tissues and paracancerous tissues were detected by qRT-PCR,the relationship between the expression level of circular RNA UBAP2 in cancer tissues and the clinical characteristics of the patients was analyzed;the patients were followed up for 3 years,the Kaplan-Meier survival curve was used to analyze the relationship between the expression level of circular RNA UBAP2 in colorectal cancer tissues and the prognosis of the patients;the multivariate COX regression was used to analyze the influencing factors of the prognosis in the patients with colorectal cancer.Results The expression level of circular RNA UBAP2 in peripheral blood of the patients with colorectal cancer was 2.06±0.97,which was significantly higher than 1.02±0.14 in the healthy controls,and the difference was statistically significant(P<0.05).Compared with the paracancerous tissues,the expression level of circular RNA UBAP2 in colorectal cancer tissues was significantly higher(P<0.05);the expression level of circular RNA UBAP2 in colorectal cancer tissues was related with the TNM stage,lymph node metastasis,invasion depth and differentiation degree(P<0.05).The Kaplan-Meier survival curve analysis showed that the 3-year survival rate of the patients with circular RNA UBAP2 high expression in colorectal cancer tissues was lower than that of the patients with low expression(P<0.05).The multivariate COX regression analysis showed that circular RNA UBAP2,TNM stage,lymph node metastasis,invasion depth and differentiation degree were the influencing factors of the prognosis in the patients with colorectal cancer(P<0.05).Conclusion Circular RNA UBAP2 has significantly high expression in peripheral blood and cancer tissues of the patients with colorectal cancer.The expression level of circular RNA UBAP2 in cancer tissues is closely related to the clinicopathological characteristics such as the TNM stage,lymph node metastasis,invasion depth and differentiation degree,and prognosis,which is of great value in judging the prognosis and survival of the patients.
