肾不藏志型不寐大鼠肾、心、脑等相关组织IL-6、TNF-α表达的差异研究

摘要 目的探讨肾不藏志型不寐大鼠与正常大鼠肾、脑、心、肝、脾、肺、大肠、膀胱组织中白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)表达的差异。方法将16只SPF级雄性SD大鼠随机分为不寐组与正常组各8只,不寐组大鼠使用D-半乳糖联合对氯苯丙氨酸构建肾不藏志型不寐模型,正常组大鼠同期正常饲养,于第46天处死大鼠后取材,免疫组化法检测2组大鼠肾、脑、心、肝、脾、肺、大肠、膀胱组织中IL-6、TNF-α表达水平。结果不寐组大鼠肾、心、脑、肺、大肠组织中IL-6表达水平和肾、脑、大肠、肺组织中TNF-α表达水平均明显高于正常组(P均<0.05),肝、脾、膀胱组织中IL-6表达水平和肝、心、脾、膀胱组织中TNF-α表达水平与正常组比较差异均无统计学意义(P均>0.05)。结论肾不藏志型不寐大鼠肾、脑、大肠、肺组织中IL-6、TNF-α表达水平及心组织中IL-6表达水平均较正常大鼠升高,这种特征性变化为从“肠-肾-脑轴”认识肾不藏志型不寐现代病理学机制提供了可能,肾不藏志型不寐大鼠肾、心、脑、肺、大肠组织中IL-6、TNF-α表达差异可能是肾不藏志型不寐睡眠相关因子的特征性变化之一。 Objective It is to investigate the differences in the expression of interleukin-6(IL-6)and tumor necrosis factor-alpha(TNF-α)in kidney,brain,heart,liver,spleen,lung,colon,and bladder tissues between rats with insomnia of type of kidney not storing the will and normal rats.Methods A total of 16 male SD rats of SPF grade were randomly divided into insomnia group and normal group,with 8 rats in each group.The rats in the insomnia model group were given D-galactose combined with p-Chlorophenylalanine by subcutaneous injection in the neck back to construct insomnia models of kidney not storing the will,the rats in the normal group were normally raised during the same period.All the rats were sacrificed on the 46th day to get the relevant visceral tissues,the expression levels of IL-6 and TNF-α in the tissues of kidney,brain,heart,liver,spleen,lung,colon,and bladder of rats in the two groups were detected by immunohistochemistry.Results The expression levels of IL-6 in the kidney,heart,brain,lung and colon tissues and the expression levels of TNF-α in the kidney,brain,colon and lung tissues of the rats in the insomnia group were significantly higher than those in the normal group(all P<0.05),and the differences in the expression levels of IL-6 in the liver,spleen and bladder tissues and the expression levels of TNF-α in the liver,heart,spleen and bladder tissues were not statistically significant between the two group(all P>0.05).Conclusion The expression levels of IL-6 and TNF-α in the kidney,brain,colon and lung tissues,and the expression level of IL-6 in the heart tissues of rats with insomnia of type of kidney not storing the will were higher than those of normal rats,and this characteristic change provides a possibility to understand the modern pathological mechanism of insomnia of type of kidney not storing the will from the“intestinal-kidney-brain axis”,and the difference in the expression levels of IL-6 and TNF-α in the kidney,heart,brain,lung,and colon tissues of rats with renal insomnia of type of kidney not storing the will could be one of the characteristic changes in the sleep-related factors of insomnia of type of kidney not storing the will.
