摘要
目的:通过对艾滋病气阴两虚证患者microRNA(miRNA)表达谱和生物信息学分析,从miRNA水平上探讨气阴两虚证的发病机制。方法:选择4例艾滋病高效抗反转录病毒治疗(HAART)后气阴两虚证患者(观察组)和4例艾滋病无典型证候者(对照组),通过高通量测序,筛选出两组之间差异表达的miRNA,进行靶基因预测和功能注释。结果:两组比较,共筛选出有显著性差异表达的miRNA 16条,其中8条上调,8条下调。差异miRNA主要参与的生物过程有多细胞生物发展、信号转导、细胞群增殖的正向调节、蛋白磷酸化等,通路主要有TNF信号通路、MAPK信号通路、酪氨酸激酶抑制剂抵抗通路。结论:艾滋病HAART后气阴两虚证与无典型证候者之间存在明显的miRNA差异性表达,中医证候的生物学基础可能与多细胞生物发展、信号转导、细胞群增殖的正向调节等相关,关键通路可能涉及TNF信号通路、MAPK信号通路等。
Objective:Through microRNA expression profile and bioinformatics analysis of AIDS patients with qi and yin deficiency syndrome,to explore the pathogenesis of qi and yin deficiency syndrome from miRNA level.Methods:Four patients with qi and yin deficiency syndrome after AIDS highly avtive anti-retroviral therapy(HAART)(observation group)and four patients with AIDS without typical syndrome(control group)were selected,through highthroughput sequencing,screened the differentially expressed miRNA between the two groups,target gene prediction and perform functional annotation.Results:Comparison of two groups,a total of 16 miRNA with significant heterosexual expression differences were screened,eight were raised and eight were lowered.Differential miRNA were mainly involved in biological processes such as multicellular biological development,signal transduction,positive regulation of cell population proliferation,and protein phosphorylation.The main pathways include TNF signaling pathway,MAPK signaling pathway and tyrosine kinase inhibitor resistance pathway.Conclusion:There is significant difference in miRNA expression between AIDS patients with qi and yin deficiency syndrome and those without typical syndrome after HAART.The biological basis of TCM syndromes may be related to the development of multicellular organisms,signal transduction,and positive regulation of cell population proliferation.The key pathways may involve TNF signaling pathway,MAPK signaling pathway,and so on.
