摘要
胡子鲇(Clarias fuscus)是我国华南地区大规模养殖的淡水鱼类。为评估胡子鲇雌雄基因组基本信息,以便为后续构建全基因组精细图谱提供基础,本研究分别选取雌雄胡子鲇进行全基因组Survey分析。根据定长核苷酸串(K-mer)分析,雌鱼和雄鱼基因组大小分别为949.69 Mb和957.96 Mb。雌鱼和雄鱼的杂合率分别为0.27%和0.23%。雌鱼和雄鱼重复序列比例分别为33.83%和33.62%。雌鱼和雄鱼基因组的G+C含量分别为39.98%和39.14%。雌鱼和雄鱼基因组组装的大小分别为926.78 Mb和992.08 Mb,雌鱼和雄鱼组装基因组N50长度分别为3082 bp和2810 bp。简单重复序列(simple sequence repeats,SSR)筛选结果显示,雌鱼和雄鱼的基因组中分别检测到1295490条和1184183条SSR序列。通过比较基因组分析,鉴定出8445条雌性特异性序列和110743条雄性特异性序列,这些性别特异性序列分别被注释到460个(雌鱼)和822个(雄鱼)基因。本研究为胡子鲇全基因组的后续构建提供了理论依据,并为后续分子育种研究提供了有价值的数据。
Hong Kong catfish(Clarias fuscus)is a widely cultured freshwater fish species in South China.In order to evaluate the basic information of male and female genomes of Clarias fuscus and provide a basis for the subsequent construction of the whole genome map,whole genome survey sequencing of male and female of Hong Kong catfish was conducted in the study.According to K-mer analysis,the genome sizes of female and male fish were 949.69 Mb and 957.96 Mb.The heterozygosity rates were 0.27%for females and 0.23%for males.The repetitive sequence rates of female and male fish were 33.83%and 33.62%,respectively.The G+C contents of the female and male genomes were 39.98%and 39.14%,respectively.The assembled genome sizes of female and male fish were 926.78 Mb and 992.08 Mb,with N50 lengths 3082 bp and 2810 bp for female and male fish,respectively.The simple sequence repeat(SSR)screening results showed that 1295490 and 1184183 SSR sequences were detected from the female and male genomes,respectively.By comparative genomic analysis,8445 female-specific and 110743 male-specific sequences were identified,and these sex-specific sequences were annotated to 460(female)and 822(male)genes,respectively.This study provides a theoretical basis for the subsequent construction of the whole genomes of Hong Kong catfish and valuable data for the subsequent molecular breeding research.
