蒙古黄芪不同组织中AmPR-10的诱导表达研究

摘要 为探究病程相关蛋白AmPR-10(Astragalus membranaceus pathogenesis-related protein-10)在蒙古黄芪生长发育中的功能,研究对试验植株分别进行盐溶液灌溉、腐皮镰孢菌侵染及水杨酸喷洒处理后,通过荧光定量PCR检测根、茎、叶中AmPR-10的表达特征。结果表明:①盐胁迫和病原菌胁迫时,根中AmPR-10的表达量与茎、叶相比为极显著(P<0.01),说明AmPR-10主要参与了根的抗胁迫过程,在响应蒙古黄芪抗病防御时发挥作用,并且在0.085 mol/L盐浓度胁迫下,根中AmPR-10的表达量在6 h出现极显著(P<0.01),腐皮镰孢菌胁迫时,第48 h也可达到极显著(P<0.01),说明AmPR-10是蒙古黄芪在遭遇逆环境时启动抵御系统且快速表达的一种蛋白;②与前2种诱导条件相比,水杨酸诱导后,茎和叶中的AmPR-10表达量明显提高,并且在5.0 mmol/L浓度下,叶片中AmPR-10表达量可在6 h达到极显著(P<0.01),茎中AmPR-10表达量也有多个极显著点(P<0.01),这说明水杨酸作为植物抗病信号分子可促进AmPR-10的表达。该研究表明,AmPR-10在蒙古黄芪生长过程中具有重要意义,为抗病黄芪幼苗的培育提供理论依据。 This paper aimed to explore the function of Astragalus membranaceus pathogenesis-related protein-10(AmPR-10)in the growth of Astragalus membranaceus.The expression characteristics of AmPR-10 in roots,stems and leaves were detected by the real-time quantitative PCR after salt solution irrigation,Fusarium solani infection,and salicylic acid spraying.The results showed that,un-der the salt stress and pathogen stress,the expression of AmPR-10 in roots was significantly higher than that in stems and leaves(P<0.01),which indicated that AmPR-10 was mainly involved in the anti-stress process of roots and could play a role in response to the disease resistance of Astragalus membranaceus.And the expression of AmPR-10 in roots showed a very significant point at 6 h(P<0.01)under the stress of 0.085 mol/L salt concentration,and it could also reach an extremely significant point at 48 h(P<0.01)un-der the Fusarium solani stress.The results demonstrated that AmPR-10 could be rapidly expressed by Astragalus membranaceus when it encountered adverse environment.Compared with above induction conditions,the expression level of AmPR-10 in stems and leaves induced by the salicylic acid was significantly increased.At the concentration of 5.0 mmol/L,the expression of AmPR-10 in leaves could reach an extremely significant point at 6 h(P<0.01),and there were several extremely significant points in stems(P<0.01).It was indicated that the salicylic acid,as a plant disease resistance signal molecule,could promote the AmPR-10 expression.It could be concluded that the AmPR-10 played an important role in the growth of Astragalus membranaceus,and provided a theoreti-cal basis for the cultivation of disease-resistant Astragalus membranaceus seedlings.
