苯酚-硫酸法测定消渴丸中多糖含量 被引量:5

摘要 目的建立消渴丸中多糖含量的测定方法。方法以葡萄糖为对照,采用苯酚-硫酸法对消渴丸中的多糖进行显色,运用紫外-可见分光光度计测定吸光度,计算消渴丸中多糖的含量,并对该方法的精密度、重复性、加样回收率等进行分析。结果该方法测得消渴丸中多糖含量在0~0.0510 mg/mL的线性范围内,相关系数为0.9999。精密度实验相对标准偏差(relative standard deviation,RSD)为0.90%,重复性实验RSD为1.38%,平均加样回收率为98.77%,RSD为3.25%。结论应用苯酚-硫酸法测定消渴丸中的多糖含量准确度高、精密度良好、重复性稳定、加样回收率高,适用于消渴丸中多糖含量的测定。 Objective To establish a method for the determination of polysaccharide content in Xiaoke pill.Methods Using glucose as a control,the phenol-sulfuric acid method was used to color the polysaccharides in Xiaoke pill,and the absorbance was measured using a UV visible spectrophotometer to determine the content of polysaccharides in Xiaoke pill.The accuracy,repeatability,and sample recovery rate of this method were analyzed.Results This method determined the content of polysaccharides in Xiaoke pill within the linear range of 0 to 0.0510 mg/mL,with a correlation coefficient of 0.9999.The relative standard deviation(RSD)of the precision experiment was 0.90%,and the RSD of repeatability experiment was 1.38%.The average sample recovery rate was 98.77%,and the RSD was 3.25%.Conclusion The application of phenol-sulfuric acid method to determine the polysaccharide content in Xiaoke pill had high accuracy,good precision,stable repeatability and high sample recovery rate,making it suitable for the determination of polysaccharide content in Xiaoke pill.
