电针对腰椎间盘突出症家兔髓核组织IL-6和血清MMP-3水平的影响

Effects of electroacupuncture on IL-6 and serum MMP-3 levels in nucleus pulposus of rabbits with lumbar disc herniation

摘要 目的:观察电针对腰椎间盘突出症(LDH)模型家兔血清中基质金属蛋白酶-3(MMP-3)及髓核组织中白介素-6(IL-6)含量的影响。方法:40只家兔随机分为空白组、模型组、穴位组和对照组,每组10只。除空白组外,均采用自制造模穿刺针造模。造模后,对穴位组和对照组家兔行电针干预,连续10 d,穴位组选取肾俞、大肠俞、环跳、委中,对照组取肾俞对照点(第2、第3腰椎棘突间旁开2 cm)、大肠俞对照点(第4、第5腰椎棘突间旁开2 cm)、环跳对照点(环跳穴上2 cm)、委中对照点(委中穴内侧旁开1 cm)。造模前后、干预后均对各组家兔综合反应能力进行评分,干预后测定各组家兔血清中MMP-3及髓核组织中IL-6含量。结果:(1)与造模前比较,造模后各组家兔综合反应能力评分均明显降低(P<0.01),家兔精神状态逐渐变差,活动减少,对外界的触觉刺激反应较迟钝,综合反应能力下降。干预后穴位组综合反应能力评分及干预前后评分差值均明显高于对照组(P<0.01),说明电针能改善家兔的综合反应能力。(2)模型组家兔髓核组织IL-6、血清MMP-3水平较空白组明显升高(P<0.01);与模型组和对照组比较,干预后穴位组家兔髓核组织IL-6、血清MMP-3表达水平明显降低(P<0.05,P<0.01),说明电针能下调髓核组织IL-6、血清MMP-3水平。结论:电针可改善LDH家兔综合反应能力,抑制炎性反应。 Objective:To observe the effects of electroacupuncture on the content of matrix metalloproteinase-3(MMP-3)in serum and interleukin-6(IL-6)in nucleus pulposus of model rabbits with lumbar disc herniation(LDH).Methods:40 rabbits were randomly divided into blank group,model group,acupoint group and control group,with 10 rabbits in each group.Except for the blank group,the self-made mold puncture needle was used to establish the model.After modeling,rabbits in acupoint group and control group were treated with electroacupuncture intervention for 10 consecutive days.In the acupoint group,Shenshu,Dachangshu,Huantiao and Weizhong were selected.In the control group,the control points of Shenshu(2 cm beside the second and third lumbar spinous processes),Dachangshu(2 cm beside the fourth and fifth lumbar spinous processes),Huantiao(2 cm above Huantiao acupoint)and Weizhong(1 cm beside the inner side of Weizhong acupoint)were selected.The integrated reaction capacity of rabbits in each group was scored before and after modeling and intervention.After intervention,the contents of MMP-3 in serum and IL-6 in nucleus pulposus were determined.Results:(1)Compared with before modeling,the integrated reaction capacity score of rabbits in all groups decreased significantly after modeling(P<0.01).The mental state of rabbits gradually deteriorated,the activity decreased,the response to external tactile stimuli was slower,and the integrated reaction capacity decreased.After intervention,the integrated reaction capacity score and the difference of score before and after intervention in the acupoint group were significantly higher than those in the control group(P<0.01),indicating that electroacupuncture could improve the integrated reaction capacity of rabbits.(2)The levels of IL-6 in nucleus pulposus and serum MMP-3 in the model group were significantly higher than those in the blank group(P<0.01);compared with the model group and the control group,the expression levels of IL-6 in nucleus pulposus and serum MMP-3 in the acupoint group were significantly decreased after intervention(P<0.05,P<0.01),indicating that electroacupuncture could down-regulate the levels of IL-6 in nucleus pulposus and serum MMP-3.Conclusion:Electroacupuncture can improve the integrated reaction capacity and inhibit inflammatory response in LDH rabbits.
