摘要
滇黄精(Polygonatum kingianum Coll. et Hemsl)是中药黄精的重要来源之一,研究采用核糖体RNA(r RNA)基因转录区ITS2序列及cp DNA非编码区rpl20-rps12、trn L-trn F和psb A-trn H序列对收集自云南省多个地区的22份滇黄精样品进行分子鉴定,并采用MEGA7.0.26构建邻接(NJ)系统聚类树,以初步了解实际流通中黄精药材的基源,探究在滇黄精药材资源利用过程中是否存在品种混淆的问题。结合各样品的分子生物学信息和形态特征,发现收集的22份样品中,初步判定可能为滇黄精的有11份,可能为卷叶黄精的有7份,其中暂无法确定为滇黄精或者卷叶黄精的有5份,鉴定为轮叶黄精的有3份,玉竹1份,多花黄精5份。这表明形态特征结合DNA分子序列分析可以作为中药材鉴定的有效手段。
Polygonatum kingianum is one of the important sources of traditional Chinese medicine Polygonatum.Through sequencing and comparison of the ribosomal RNA(rRNA)gene transcription region ITS2 sequence and cpDNA non-coding regions rpl20-rps12,trnL-trnF and psbA-trnH sequences,we performed molecular identification for 22 samples from multiple regions of Yunnan Province.MEGA7.0.26 was used to construct the cluster tree of the adjacent(NJ)system to preliminarily understand the origin of Polygonatum materials in actual circulation and to explore whether there was variety confusion in the process of utilization of the medicinal resource Polygonatum kingianum.According to the molecular biological information and morphological characteristics,of the 22 samples collected 11 samples were preliminarily identified as Polygonatum kingianum,seven Polygonatum cirrhifolium,five unidentified,three Polygonatum verticillatum,one Polygonatum odoratum,and five Polygonatum cyrtonema.This shows that combining morphological features with DNA molecular sequence analysis can be an effective method for identifying traditional Chinese medicinal materials.
