基于SSR标记的不同粒色大麦品种(系)的遗传差异分析

摘要 【目的】研究不同粒色大麦品种(系)的遗传差异,为有色籽粒大麦的品种选育奠定基础。【方法】基于大麦7条染色体上的51个简单重复序列(simple sequence repeats,SSR)标记位点,分析25份不同粒色大麦品种(系)的遗传差异。【结果】在25份参试品种(系)基因组中,51个SSR位点发生2~16个等位变异,PIC值变幅为0.491~0.922。品种(系)间的遗传相似系数变幅为0.569~0.872,其中wn6与wn16的遗传相似系数最小,wn16与wn17的遗传相似系数最大。聚类分析结果显示:3个黑色籽粒品系(wn-16、wn-17、wn-18)聚为第1类,3个紫黑色籽粒品种(wn-13、wn-14、wn-15)和6个蓝色籽粒品种(wn-20、wn-21、wn-22、wn-23、wn-24、wn-25)聚为第2类,5个褐色籽粒品种(wn-1、wn-2、wn-3、wn-4、wn-5)、3个黄色籽粒品种(wn-6、wn-7、wn-8)和4个紫色籽粒品种(wn-9、wn-10、wn-11、wn-12)聚为第3类。【结论】相同粒色的大麦品种(系)具有较高的遗传相似性,wn-6与wn-16、wn-6与wn-18、wn-1与wn-24等配对品种(系)的遗传相似系数较低,可用作有色籽粒大麦品种选育的杂交亲本。 [Purpose]To study the genetic differences among barley varieties(lines)with different grain colors,laying a foundation for the breeding of special varieties in flavonoids.[Methods]A total of 51 simple sequence repeats(SSR)loci distributed in seven chromosomes of barley were used to analyze the genetic differences among barley varieties(lines)with different grain colors.[Results]The number of allele mutants in different SSR loci ranged from 2 to 16 in 25 material genomes,the polymorphism information content(PIC)of the different SSR loci ranged from 0.491 to 0.922.The genetic similarity coefficients among 25 barley varieties ranged from 0.569 to 0.872,with the smallest genetic similarity coefficient between wn-6 and wn-16,and the largest genetic similarity coefficient between wn-16 and wn-17.The results of cluster analysis showed that three black grain lines(wn-16,wn-17 and wn-18)were clustered into the first group,three purple grain varieties(wn-13,wn-14 and wn-15)and six blue grain varieties(wn-20,wn-21,wn-22,wn-23,wn-24 and wn-25)were clustered into the second group,and five brown grain varieties(wn-1,wn-2,wn-3,wn-4 and wn-5),three yellow grain varieties(wn-6,wn-7 and wn-8)and four purple grain varieties(wn-9,wn-10,wn-11 and wn-12)were clustered into the third group.[Conclusion]The barley varieties(lines)with the same grain color show relatively high genetic similarity,and the genetic similarity between wn-6 and wn-16,wn-6 and wn-18,wn-1 and wn-24 are relatively lower,the paired materials can be used as hybrid parents for barley breeding specially for flavonoids.
