富勒醇促进小鼠皮肤创面愈合的疗效及机制

摘要 目的探究生物材料富勒醇在小鼠皮肤创面愈合中的疗效及机制。方法将24只6~8周龄雄性C57BL/6小鼠随机分为假手术组(对照组)、模型组+生理盐水组(NS组)及模型组+富勒醇组(FUL组),每组8只。FUL组在建模后腹腔注射10 mg·kg^(-1)·d^(-1)富勒醇7 d,NS组及对照组注射等体积生理盐水。每日记录小鼠体质量,分别于治疗第3、7天对创面进行拍照并计算相应的创面愈合率。治疗后通过HE染色和Masson染色观察创面皮肤组织愈合情况;RT-qPCR检测创缘组织TNF-α、IL-6、IL-1βmRNA水平;Western blot检测创缘组织NF-κB、TNF-α、IL-6、IL-1β蛋白水平;ELISA检测小鼠血清TNF-α、IL-6、IL-1β及创缘组织MDA、SOD水平。结果各组小鼠每天的体质量无明显变化(P>0.05)。第3、7天,FUL组创面愈合率明显高于NS组(P<0.05);与NS组相比,干预结束时FUL组创缘组织NF-κB、IL-1β、IL-6、TNF-α及MDA水平明显下降(P<0.05),而SOD水平显著升高(P<0.05),同时血清中IL-1β、IL-6、TNF-α表达明显下降(P<0.05)。HE染色及Masson染色显示,与NS组相比,FUL组上皮结构连续、完整,胶原纤维更加密集且排列整齐,上皮爬行距离更远(P<0.05),肉芽组织更厚(P<0.05),胶原容积分数更高(P<0.05)。结论富勒醇可以有效促进小鼠皮肤创面愈合,其可能是通过抑制NF-κB信号通路及下游炎症因子表达实现的。 Objective To explore the efficacy and mechanism of the biomaterial fullerenol(FUL)on skin wound healing of mice.Methods A total of 24 male C57BL/6 mice aged from 6 to 8 weeks were randomly divided into the sham operation group(control group),model group+normal saline group(NS group)and the model group+fullerenol group(FUL group),with 8 mice in each group.Mice in the FUL group injected intraperitoneally with 10 mg·kg^(-1)·d^(-1)fullerenol for 7 days,and mice in the NS group and the control group injected with an equal volume of normal saline.The daily body weight of mice were recorded,and the wounds were photographed on the 3rd and 7th days of treatment to calculate the corresponding wound healing rate.The wound skin tissue healing after treatment was observed by HE and Masson stainings,the mRNA levels of TNF-α,IL-6,IL-1βin wound margin tissue were detected by RT-qPCR,the protein levels of NF-κB,TNF-α,IL-6,IL-1βin wound margin tissue were detected by Western blot,the levels of TNF-α,IL-6,IL-1βin serum and MDA,SOD in wound margin tissue were detected by ELISA.Results There was no significant change in daily body weight among groups(P>0.05).On the 3rd and 7th days,the wound healing rate of the FUL group was significantly higher than that of the NS group(P<0.05).Compared with the NS group,the levels of NF-κB,IL-1β,IL-6,TNF-αand MDA in the wound margin tissue of the FUL group were significantly decreased(P<0.05),while the level of SOD was significantly increased(P<0.05),and the expression of serum IL-1β,IL-6,and TNF-αwere significantly decreased(P<0.05).The HE staining and Masson staining showed that the FUL group had a continuous and complete epithelial structure,denser and neatly arranged collagen fibers,longer epithelial crawl distance(P<0.05),thicker granulation tissue(P<0.05),and higher collagen volume fraction compared with the NS group(P<0.05).Conclusion Fullerenol can effectively promote the skin wound healing of mice,which may be achieved by inhibiting the NF-κB signaling pathway and the expression of downstream inflammatory factors.
