摘要
目的:提高实验室对食品中致泻大肠埃希氏菌(Diarrheagenic Escherichia coli,DEC)的检测能力与检测效率,验证DEC多重PCR试剂盒的适用性。方法:依据《食品安全国家标准食品微生物学检验致泻大肠埃希氏菌检验》(GB 4789.6—2016)分离鉴定样品23-H770、23-X371,用DEC多重PCR检测试剂盒检测5种DEC的12条特征基因。结果:23-H770未检出DEC,23-X371检出STEC/EHEC,阴、阳性对照菌种均检出GB 4789.6—2016中5种DEC目标条带与型别对照表相应基因条带,且空白对照12条目标基因条带均未检出。结论:能力验证样品评价结果为“满意”,多重PCR检测试剂盒能快速准确检测EPEC、EIEC、ETEC、STEC/EHEC和EAEC。
Objective:To improve the detection capability and efficiency of diarrheagenic Escherichia coli(DEC)in food,and to verify the applicability of DEC multiplex PCR kit.Method:Samples 23-H770 and 23-X371 were isolated and identified according to GB 4789.6—2016,and 12 characteristic genes of 5 species of DEC were detected with DEC multiple PCR detection kit.Result:DEC was not detected in 23-H770,STEC/EHEC was detected in 23-X371,and GB 4789.6—2016 were detected in all the five DEC target bands and corresponding gene bands in the genotype comparison table,while none of the 12 target gene bands were detected in the blank control.Conclusion:The evaluation result of ability verification sample is satisfactory.The multiplex PCR kit can detect EPEC,EIEC,ETEC,STEC/EHEC and EAEC quickly and accurately.
