桃核承气汤对慢性肾衰竭大鼠Wnt4/β-catenin信号通路中p-GSK-3β的影响

Effect of Taohe Chengqi Decoction on p⁃GSK⁃3βin Rats with Chronic Renal Failure Based on Wnt4β⁃catenin Signal Path

目的探讨慢性肾衰竭(CRF)大鼠肾纤维化(RIF)过程中,桃核承气汤对Wnt4/β-catenin信号通路p-GSK-3β的影响。方法100只清洁级Wistar大鼠随机分为正常组、假手术组、模型组、治疗组,各25只,模型组和治疗组行5/6肾切除术,假手术组仅做双肾被膜剥离术,正常组不做处理。造模成功后,治疗组予桃核承气汤灌胃,其余3组予等量生理盐水灌胃,给药时间共为8周。Western Blot法检测肾组织中Wnt4/β-catenin信号通路的相关蛋白含量,光镜观察肾组织。结果Wnt4、p-GSK-3β、β-catenin蛋白含量,治疗组比模型组显著降低(P<0.05);光镜观察,模型组的肾小球结构固缩,肾间质纤维增生严重,治疗组明显减轻。结论桃核承气汤对CRF的作用机制,可能与降低Wnt4、p-GSK-3β、β-catenin蛋白含量从而调节Wnt4/β-catenin信号通路有关。

Objective To discuss the influence of Taohe Chengqi(THCQ)decoction on p⁃GSK⁃3βregulation mechanism in renal fibrosis of rats with chronic renal failure based on Wnt4/β⁃catenin signal path.Methods One hundred clean Wistar rats were randomly divided into normal group,pseudo operation group,model group and treatment group.The model and treatment group received 5/6 nephrectomy.The treatment group was given THCQ decoction by gavage.Related protein content of Wnt4/β⁃catenin signal pathway were checked by Western Blot.Renal tissue was observed by light microscope.Results Compared with the model group,the protein content of Wnt4,p⁃GSK⁃3βandβ⁃catenin in the treatment group significantly decreased(P<0.05).The results of light microscope observation showed that the degree of glomerulosclerosis and renal interstitial fibrosis in the treatment group was significantly less than that in the model group.Conclusion The mechanism of THCQ decoction on CRF related to Wnt4/β⁃catenin signal pathway,which might be adjusted by the protein content decrease of Wnt4,p⁃GSK⁃3βandβ⁃catenin.
