基于氧化石墨烯的无酶循环放大荧光信号法检测DNA

摘要 基于纳米材料氧化石墨烯的荧光猝灭性能,引入靶物催化发卡结构组装作为循环信号放大方法,构建了一种用于检测单链DNA的高灵敏荧光检测平台.通过对实验条件进行优化,确定以氧化石墨烯浓度60 mg/mL、加入target DNA后反应温度37℃、反应时间30 min为该检测方法的最佳反应条件.在优化后的实验条件下,加入靶物DNA后荧光信号变化值与靶物浓度的线性范围为0.5~14 pM,检测限为0.055 pM.该检测方法快速、灵敏、成本低,通过改变荧光探针序列还可实现micro RNA、生物小分子和蛋白质的检测,因此在生化分析和分子医学等领域具有较大的应用前景. Based on the fluorescence quenching performance of nanomaterials graphene oxide,a highly sensitive fluorescence detection platform for detecting single strand DNA was constructed by introducing the target catalytic hairpin structure assembly as a cyclic signal amplification method.By optimizing the experimental conditions,it was determined that the concentration of graphene oxide was 60 mg/mL,the reaction temperature was 37 ℃,and the reaction time was 30 min after adding target DNA as the optimal reaction conditions.Under the optimized experimental conditions,the linear range of fluorescence signal change value and target concentration after adding target DNA was 0.5~14 pM,and the detection limit was 0.055 pM.The detection method is fast,sensitive and low-cost,and can also detect microRNA,biological small molecules and proteins by changing the fluorescent probe sequence.Therefore this detection method has great application prospects in biochemical analysis and molecular medicine.
