莪术-三棱药对醋制前后指纹图谱的建立及多指标成分含量变化

摘要 目的:建立莪术-三棱药对醋制前后HPLC指纹图谱,对5种指标性成分进行定量分析,探索莪术-三棱药对醋制前后的质量差异。方法:采用HPLC法建立莪术-三棱药对醋制前后指纹图谱,通过化学计量学方法对莪术-三棱药对醋制前后指纹图谱进行分析;并采用HPLC法测定莪术-三棱药对醋制前后对羟基苯甲酸、对羟基苯甲醛、香兰素、姜黄素、莪术醇的含量。结果:结果表明20批样品可以明显的分为2类,莪术-三棱生品与醋品在整体化学成分上具有一定的差异。含量测定结果表明,莪术-三棱药对经过醋制后,指标成分含量均有不同程度的增加,其中对羟基苯甲酸、对羟基苯甲醛、香兰素、姜黄素、莪术醇的平均含量增加了11.11%、36.31%、30.18%、11.68%、53.43%。结论:所建立的指纹图谱及多指标含量测定方法稳定可靠,从定性与定量评价了莪术-三棱药对醋制前后的质量差异,为莪术-三棱药对炮制研究提供一定的科学依据。 Objective:To establish an HPLC fingerprint of Ezhu-Sanleng medicine pair before and after vinegar preparation,quantitatively analyze 5 indicator components,and explore the quality differences of Ezhu-Sanleng medicine pair before and after vinegar preparation.Method:HPLC method was used to establish the fingerprint of Ezhu-Sanleng medicine pair before and after vinegar processing,and chemometric method was used to analyze the fingerprint of Ezhu-Sanleng medicine pair before and after vinegar processing.The contents of p-hydroxybenzoic acid,p-hydroxybenzaldehyde,vanillin,curcumin and curcumol were determined by HPLC before and after the vinegar preparation of Ezhu-Sanleng medicine pair.Result:The results indicated that 20 batches of samples can be clearly divided into 2 categories,and there were certain differences in overall chemical composition between the raw and vinegar products of Ezhu-Sanleng medicine pair.The content determination results showed that the contents of the index components of Ezhu-Sanleng medicine pair increased in varying degrees after vinegar processing,among which the average content of p-hydroxybenzoic acid,p-hydroxybenzaldehyde,vanillin,curcumin,and curcumol increased by 11.11%,36.31%,30.18%,11.68%and 53.43%,respectively.Conclusion:The established fingerprint and multi indicator content determination method are stable and reliable.The qualitative and quantitative evaluation of the quality differences before and after vinegar preparation of Ezhu-Sanleng medicine pair is conducted to provide a certain scientific basis for the research on the processing ofEzhu-Sanleng medicine pair.
