摘要
目的:探讨漆赛尔桑当松汤对人体类风湿关节炎成纤维样滑膜细胞(HFLS-RA)中NOTCH和TLR4mRNA相对表达量的影响。方法:采用逆转录-聚合酶链反应(RT-PCR)检测TNF-α诱导的HFLS-RA中NOTCH1、NOTCH3和TLR4mRNA表达。分为空白组、模型组、漆赛尔桑当松汤含药血清组。结果:造模后NOTCH1、NOTCH3、TLR4的mRNA相对表达量与空白组相比有上升(P<0.05);治疗后漆赛尔桑当松汤含药血清组中的NOTCH1、NOTCH3、TLR4有降低趋势,且有显著性差异(P<0.05)。结论:漆赛尔桑当松汤能抑制TNF-α诱导的HFLS-RA中NOTCH和TLR4基因的表达。
Objective:To investigate the effects of Qi-Sai-Er-Sang-Dang-Song Decoction on the mRNA relative expression of NOTCH and TLR4 genes in human fibroblastic synoviocytes-rheumatoid arthritis(HFLS-RA).Method:The mRNA relative expression of NOTCH1,NOTCH3,and TLR4 in TNF-α-induced HFLS-RA was detected by reverse transcription-polymerase chain reaction(RT-PCR).The experiment was divided into control group,model group,Qi-Sai-Er-Sang-Dang-Song Decoction drug-containing serum group.Result:Compare to control group,the mRNA relative expression of NOTCH1,NOTCH3,and TLR4 was increased after TNF-α-induced HFLS-RA(P<0.05),while the mRNA relative expression was decreased after administering the drug-containing serum(P<0.05).Conclusion:Qi-Sai-Er-Sang-Dang-Song Decoction can suppress the mRNA relative expression of NOTCH and TLR4 in TNF-α-induced HFLS-RA.
