摘要
目的:通过对提取过程各阶段产物的质量评价实现疏肝利胆丸提取工艺方法的优化与条件优选。方法:采用L_(9)(3^(4))正交试验方法,利用水蒸气蒸馏法提取挥发油,高效液相色谱法测定干浸膏中黄芩苷含量,分别以挥发油提取量、浸膏得率和黄芩苷提取量为指标,考察浸泡时间、加水量、提取时间等因素对指标提取效果的影响。结果:优选的挥发油提取工艺为加10倍量水,浸泡1 h,提取挥发油5 h,所得挥发油量大于5 mL。优选的水提优选工艺为1.97倍水浸泡1 h,加水量8倍,提取2 h,提取1次,干浸膏中黄芩苷含量大于0.59%。结论:本研究优选的提取工艺稳定可行、效率高,可用于疏肝利胆丸的提取,并为该丸剂的质量控制提供重要的理论依据。
OBJECTIVE This study aimed to optimize the extraction process and select the optimal conditions for the extraction of Shugan Lidan Pills by evaluating the quality of the products at each stage of the extraction process.METHODS Volatile oil was extracted using steam distillation,and the baicalin content in the dried extract was determined using high-performance liquid chromatography.The L_(9)(3^(4))orthogonal experimental method was used to investigate the effects of factors such as soaking time,amount of added water,and extraction time on the extraction efficiency of the active ingredients,with the extraction yield of volatile oil,yield of dried extract,and content of baicalin as the evaluation indices.RESULTS The optimized extraction process for volatile oils wasto add 10 times the amount of water,soak for 1 hour,and extract the volatile oils for 5 hours,resulting in a volatile oil yield of more than 5 mL.The optimal water extraction process was to soak in 1.97 times the amount of water for 1 hour,add 8 times the amount of water,extract for 2 hours,and perform 1 extraction,with a baicalin content in the dried paste greater than 0.59%.CONCLUSION The optimized extraction process in this study is stable,feasible,and efficient,and can be used for the extraction of Shugan Lidan Pills.It provides an important theoretical basis for quality control of this formulation.
