LncRNA-ANCR对骨肉瘤细胞生物学行为的影响

摘要 目的:探讨骨肉瘤细胞中LncRNA-ANCR的表达情况以及LncRNA-ANCR对骨肉瘤细胞生物学的影响。方法:采用荧光定量PCR检测实验各组细胞中LncRNA-ANCR的表达水平,采用CCK-8法检测各组细胞的增殖率,采用流式细胞术检测细胞凋亡,采用Transwell小室法检测细胞的侵袭、迁移能力。结果:MG-63和UMR-106骨肉瘤细胞LncRNA-ANCR的表达水平最高,选择其进行后续实验研究;与WT组相比较,Mock组、NC组细胞中LncRNA-ANCR的表达水平、转染后细胞的增殖率、细胞凋亡率、侵袭细胞和迁移数均无明显变化(P>0.05);与WT组相比较,siRNA组细胞中LncRNA-ANCR的表达水平、转染后增殖率、细胞的侵袭和迁移数显著下降(P<0.05),细胞的凋亡率明显上升(P<0.05)。结论:LncRNA-ANCR可促进MG-63和UMR-106骨肉瘤细胞的增殖、侵袭以及转移,抑制细胞的凋亡。 Objective:To investigate the expression of LncRNA-ANCR in osteosarcoma cells and the effect of LncRNA-ANCR on the biology of osteosarcoma cells.Methods:The expression level of LncRNA-ANCR in each group was detecteol by fluorescence quantitative PCR.The proliferation rate of each group was detected by CCK-8 method.Flow cytometry was used to detect apoptosis.Transwell chamber method was used to detect cell invasion and migration ability.Results:The expression level of LncRNA-ANCR in MG-63 and UMR-106 osteosarcoma cells was the highest,and it was selected for subsequent experimental research.Compared with the WT group,there was no significant change in the expression level of LncRNA-ANCR,cell proliferation rate,apoptosis rate,invasive cells and migration number in Mock group and NC group(P>0.05).Compared with the WT group,the expression level of LncRNA-ANCR,the proliferation rate after transfection,the number of invasion and migration of cells decreased significantly(P<0.05),and the apoptosis rate of cells increased significantly in the siRNA group(P<0.05).Conclusion:LncRNA-ANCR can promote the proliferation,invasion and metastasis of MG-63 and UMR-106 osteosarcoma cells,and inhibit cell apoptosis.
