HC2作为急性缺血性脑卒中诊断的潜在生物标志物

Heparin Cofactor Ⅱ Is a Potential Biomarker for the Diagnosis of Acute Ischemic Stroke

摘要 为了探究急性缺血性脑卒中(Acute ischemic stroke,AIS)患者和对照组静脉血血清中的O-GlcNAc糖蛋白差异,筛选AIS诊断的潜在生物标志物.首先对AIS和对照组血清中的O-GlcNAc糖基化修饰进行检测,发现AIS的O-GlcNAc修饰程度明显高于对照组(P<0.05),通过CpOGAD298N富集AIS和对照组血清中的OGlcNAc蛋白并进行质谱鉴定,得到差异蛋白肝素辅助因子Ⅱ(Heparin Cofactor Ⅱ,HC2),Western blot检测结果表明,富集蛋白中HC2丰度在AIS组显著高于对照组(P<0.01),ROC曲线分析显示,HC2对急性缺血性脑卒中具有潜在的诊断价值(AUC=0.882),并且高于常用的血脂指示标志. To explore the difference between O-GlcNAcylated proteins in the serum of acute ischemic stroke(AIS)patients and the control group,and to screen the potential biomarkers for AIS diagnosis,the OGlcNAcylation in the serum of AIS and the control group was detected.The O-GlcNAcylation level in AIS was significantly higher than that in the control group(P<0.05).CpOGA(D298N)was used to enrich OGlcNAcylated proteins in the serum,and the differential protein HC2 was confirmed by mass spectra analyses and western blot.It was found that the amount of enriched HC2 in AIS was significantly higher than that in the control group(P<0.01).The enriched HC2 has a potential diagnostic value for acute ischemic stroke(AUC=0.882),which is better than common blood lipid indicators.
