NFKB1基因多态性与冠心病(心血瘀阻证)及炎症因子的相关性研究

摘要 目的 探讨NFKB1启动子区-94ins/del ATTG(rs28362491)与冠心病(心血瘀阻证)及炎症因子的关系。方法 选取冠心病患者173例,根据中医辨证分型,分为心血瘀阻证69例和非心血瘀阻证104例,同期非冠心病患者107例为对照组。采用聚合酶链反应-限制性片段长度多态性技术分析NFKB1-rs28362491基因多态性,检测肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)水平。结果 冠心病(心血瘀阻证)NFKB1基因多态性位点DD基因型及D等位频率明显高于冠心病非心血瘀阻证及对照组(P <0.05);冠心病(心血瘀阻证)患者血清TNF-α、IL-6、IL-8水平明显高于冠心病非心血瘀阻证及对照组(P <0.05);DD基因型组TNF-α、IL-6、IL-8水平均高于ID和II基因型组(P <0.05)。结论 NFKB1-rs28362491基因多态性与冠心病(心血瘀阻证)有关联,DD基因型可能干预TNF-α、IL-6、IL-8水平成为冠心病(心血瘀阻证)易感性遗传因素。 Objective:To investigate the association of NFKB1 promoter region-94ins/del ATTG(rs28362491)of coronary heart disease(syndrome of blockade due to heart blood stasis)and inflammatory factors.Methods:A total of 173 patients with coronary heart disease were enrolled.According to TCM syndrome differentiation,they were divided into 69 patients with syndrome of blockade due to heart blood stasis and 104 patients with non syndrome of blockade due to heart blood stasis;another 107 non coronary heart disease patients in the same period were served as the control group.NFKB1-rs28362491 gene polymorphism was analyzed by poly⁃merase chain reaction-restriction fragment length polymorphism technique,and TNF-α,IL-6 and IL-8 levels were detected.Results:NFKB1 polymorphisms in the DD genotype and D allele frequency were significantly high⁃er in patients with syndrome of blockade due to heart blood stasis than non-syndrome of blockade due to heart blood stasis and control group(P<0.05);serum levels of TNF-α,IL-6,and IL-8 were significantly higher in patients with syndrome of blockade due to heart blood stasis than non-syndrome of blockade due to heart blood stasis and control group(P<0.05);the levels of TNF-α,IL-6 and IL-8 in DD genotype were higher than those in ID and II genotype(P<0.05).Conclusion:The NFKB1-rs28362491 gene polymorphism is associated with coro⁃nary heart disease(syndrome of blockade due to heart blood stasis),and the DD genotype may interfere with TNFα,IL-6,and IL-8 levels as genetic predisposing factors for coronary heart disease(syndrome of blockade due to heart blood stasis).
