维甲酸结合蛋白CRABPs在肿瘤中的作用研究进展

摘要 维甲酸(Retinoic acid,RA)是维生素A的衍生物,具有疏水性,需与脂质结合蛋白家族的成员结合而进行细胞运输。脂质结合蛋白包括细胞视黄酸结合蛋白1(cellular retinoic acid binding protein 1,CRABP1)和细胞视黄酸结合蛋白2(CRABP2)及脂肪酸结合蛋白5(fatty acid-binding protein,FABP5)。CRABPs在胚胎中广泛表达,但它们通常不会共存于同一细胞中,表明两种CRABP在RA中具有不同的生物学功能。RA通过调节细胞增殖,分化和死亡在发育过程中起着重要作用。近年来研究表明CRABPs在正常成人组织中表达低,而在各类肿瘤中异常升高,但却不共同表达,提示二者在肿瘤进展发挥不同作用。本文综述了CRABP1和CRABP2在各类肿瘤中的研究进展,并总结了CRABPs参与肿瘤发展的作用与分子机制。 Retinoic acid(RA)is a derivative of vitamin A,which is hydrophobic and responsible for cell transport after binding to lipid-binding proteins.The latter includes cellular retinoic acid binding protein 1(CRABP1),cell retinoic acid binding protein 2(CRABP2)and fatty acid binding protein 5(FABP5).The cellular retinoic acid-binding proteins(CRABPs)are widely expressed in embryos,but they are usually not co-expressed in the same type of cells,indicating that the two CRABPs have different biological functions involved in RA transportation and cell functions.RA is an important signal molecule that regulates the developmental process via mediating cell proliferation,differentiation and death.Recent studies have shown that CRABPs are lowly expressed in tissues of healthy adults,but abnormally overexpressed in tumors.They are not co-expressed,suggesting the different roles in tumor progression.This review describes the research progress of CRABP1 and CRABP2 in tumors,and summarizes their function and mechanism in tumor progression.
